芪参益气滴丸对Ameroid缩窄环致心功能不全小型猪心肌细胞凋亡及相关蛋白表达的影响

摘　　要：目的:观察芪参益气滴丸对Ameroid缩窄环致心功能不全小型猪心肌细胞凋亡及相关蛋白表达的影响。方法:巴马小型猪22只,采用左冠状动脉前降支放置缩窄环的方式制备小型猪慢性心肌缺血心功能不全模型,随机分为模型组、地高辛组、芪参益气滴丸组,每组6只,另制备假手术动物4只。于术后第4周开始口服给药,地高辛组每日1次,每次0.0125mg·kg-1,芪参益气滴丸组每日3次,每次0.025g·kg-1,持续8周。于给药后8周末取材,采用TUNEL法进行心肌组织细胞凋亡率检测,采用Western-blot法检测TNF1、TNFR、Active Caspase-3及Caspase-8表达情况。结果:TUNEL结果显示,模型组心肌组织细胞凋亡率与假手术组比较显著升高,芪参益气滴丸组心肌组织细胞凋亡率与模型组比较显著降低。Western-blot结果显示,模型组梗死边缘区心肌组织TNF-α、Active Caspase-3含量显著升高,芪参益气滴丸组TNF-α、Active Caspase-3含量显著下降。结论:芪参益气滴丸能够显著抑制Ameroid缩窄环致心功能不全小型猪心肌细胞凋亡,其作用机制可能与抑制TNF-α和active Caspase-3表达有关。Objective： To study the effect of Qishen Yiqi Dropping Pills on apoptosis and related protein in Ameroid narrow ring caused heart failure of miniature pigs. Methods ： Animal models of heart failure were induced by Ameroid nar- row ring,and were randomly divided into model group,digoxin group and Qishen Yiqi Dropping Pill group with 6 in each group, sham operation group with 4. Oral administration was began at 4 weeks after surgery, digoxin group used digoxin O. 0125 mg kg-1 ,once a day. Qishen Yiqi Dropping Pill group used the corresponding drug O. 025g kg-1 ,3 times a day ,for 8 weeks. TUNEL was used to evaluate the apoptosis rate ,and Western - blot was used to evaluate the expressions of TNF-α, TNFR, active Caspase -3 and Caspase- 8 in each group of animals at the end of 8th week after administra- tion. Results： TUNEL experiments showed that myocardial apoptosis was significantly lower than that of the model group in Qishen Yiqi Dropping Pill group. Western -blot showed that TNF-α and active Caspase -3 levels were significantly increased in myocardial infarct marginal zone in model group, and TNF -α as well as active Caspase -3 was significantly decreased in Qishen Yiqi Dropping Pill. Conclusion： Qishen Yiqi Dropping Pill can protect the myocardium from apopto- sis by regulating the TNF-α and active Caspase -3 levels.
