摘 要:目的 利用RP-HPLC法对金银花、黄芩单煎液和混煎液进行分析。方法 色谱柱:Phenomenex C18(250 X 4.6nm,5μm),流动相:甲醇与2%磷酸溶液进行梯度洗脱,检测波长为322nm,柱温:25℃;流速:1.0mL/min;进样量:5μL。结果 绿原酸和黄芩苷分别在下列范围内具有良好的线性关系:0.21~4.2μg(r=0.999 9);0.155 2~3.104μg(r=0.999 9)。回收率(n=6)分别为99.61%(RSD 2.5%),102.76%(RSD 2.1%)。对银花-黄芩药对的色谱峰的来源进行了归属,测定了绿原酸和黄芩苷的含量,其在单煎液和混煎液中的含量差异无统计学意义。结论 银花-黄芩药对的物质基础和金银花、黄芩单用的物质基础有异同,为临床和制药提供了实验依据,同时也为中药复方研究提供了参考,具有一定的理论和实践意义。Objective To analyse single and mixed decoction of honeysuckle and scutellaria by RP--HPLC.Methods Phenomenex CI8 (250 × 4.6nm, 5μm) column was used with mixture of methanol and 2% phosphoricacid in a gradient mode as mobile phase, the wavelength of measurement was 322 nm, 1.0mL/min as flow-rate.The column temperature was set at 25 ℃. Results The linear ranges of chlorogenic acid and baicalin were 0.21-4.2μg(r=0.999 9); 0.155 2-3.104 μg(r=0.999 9), respectively. The average recoveries of chlorogenic acid and baicalinwere 99.61% (n=6, RSD 2.5%) and 102.76% (n=6, RSD 2.1% ), respectively. Major peaks were confirmed to thetwo component herbs, contents of chlorogenic acid and baicalin were determinated, no statistical significance wasobtained in the concentration of chlorogenic acid and baicalin between the single decoction, mixed decoction.Conclusion RP--HPLC separations showed that there was difference among the single decoction and mixeddecoction,which provide increasing experimental evidence for clinic medicine and pharmaceutics and suppliedreference for the complex prescription of traditional Chinese medicine.
