肥胖大鼠尿液足糖萼蛋白的检测及意义

摘　　要：目的:检测高脂饮食诱导的肥胖大鼠尿液足糖萼蛋白(Podocalyxin,PCX)的变化并探讨其临床意义。方法:Wistar大鼠分为正常对照组(Con组)和肥胖组(Ob组)。在造模14周时,分别用光学显微镜、电子显微镜观察肾脏形态变化,用ELISA方法检测尿PCX,由生化自动分析仪测定视黄醇结合蛋白(Retinol conjugated protein,RBP)、尿微量白蛋白。结果:(1)0周时Ob组与Con组比较性别、体重、身长、尾长、Lee's指数等无差异,14周时Ob组体重、身长、Lee's指数明显高于Con组(P<0.05)。(2)14周时Ob组与Con组比较RBP无明显差异,Ob组尿微量白蛋白明显高于Con组(P<0.05)。(3)Ob组在14周时尿PCX排泄明显多于Con组。(4)光镜下Ob组大鼠肾小球变化不明显。电镜下Ob组大鼠肾脏肾小球足细胞呈粗大状,足突轻度微绒毛化。结论:肥胖大鼠尿液PCX排泄增多,有望成为肥胖导致肾损害的标志物。Objective： To test the expression of urine Podocalyxin （PCX） in obesity rats induced by high fat diet. Methods： The rats were divided into two groups ： normal control group （ Con group） and obesity group （ Ob group）. Both of the group rats had been bred for 14 weeks. The tissue structure of the rats kidneys was studied under light microscope and transmission electron microscope. The expression of urine PCX was measured by ELISA. The urine retinal binding protein （RBP） and microalbumin were tested by automatic biochemistry analyzer. Results： （1） There was no difference in gender,weight,body long and Lee＇s index between the two groups at beginning. But after 14 weeks, the weight, body long and Lee＇s index of Ob group were higher than those of Con group （P〈0. 05）. （2） The urine RBP was no significant difference between the two groups. The urine microalbumin was increased in Ob group（P〈0. 05）. （3） The urine PCX protein of Ob group was higher than that of Con group （P〈0.05）. （4）There was no change in the glomerular structure of Ob rats after under light microscope. The podocyte of the kidney of Ob was observed to become broader and villus-like under transmission electron microscope. Conclusion： The urine PCX is increased in Ob group. It may be a marker of obesity related kidney damage.
