摘 要:目的:分析ITP患者外周血单个核细胞来源树突状细胞(MoDC)的活化状态,以进一步明确免疫性血小板减少症(ITP)患者树突状细胞(DC)存在异常的机制。方法:将ITP患者分为初诊组、难治组、有效组(完全反应+有效),以正常健康者为对照组。流式细胞仪检测各组MoDC活化前吸收葡聚糖(dextran)抗原能力(MFI),流式细胞仪检测各组MoDC活化后细胞表面CD80、CD86的表达率以确定DC分化、成熟能力,ELISA法检测各组MoDC活化培养上清的IL-6、IL-12的含量。结果:难治组、初治组MFI明显高于对照组及有效组,难治组、初治组的CD86、CD80表达均明显高于对照组及有效组,难治组、初治组的IL-6、IL-12均明显高于对照组及有效组,有效组的IL-6、IL-12均明显高于对照组。结论:ITP患者未活化MoDC吸收抗原能力增强,活化后分化成熟度较高,这些可能与ITP的发病机制有关。Objective:Defective dendritic cells (DCs) functions have been implicated in ITP, to investigate the activation of DCs in immune thrombocytopenia (ITP) patients. Methods: ITP patients were studied before treatment (group untreated ) , after treatment with improvement (group improvement ) and after treatment with refractory( group refractory), the antigen dextran uptake by immature MoDC was tested with flow cytometry. The levels of CD80, CD86 on the surface of activated MoDC was also tested with flow cytometry. The levels of IL-6, IL-12 in supernatant of activated MoDC in different group were tested with ELISA. Results: The MFI (mean florescent intensity) of antigen uptake by immature MoDC in group refractory and group untreated was higher than that in group control or group improvement, the level of CD86 , CD80 of activated MoDC in group refractory and group untreated were higher than that in group control or group improvement. The levels of IL-6,IL-12 in group refractory and group untreated were higher than that in group control or group improvement obviously (P〈0. 01 ). Conclusion: In ITP, the immature MoDCs has stronger antigen-uptake ability, the activated MoDC has more matured activity, these may play an important role in the pathogenesis of ITP.
