摘 要:目的:观察益气化瘀解毒方对人肝癌移植瘤及HIF1a、E-cad表达的影响。方法:建立人肝癌移植瘤模型,分为7组,分别以益气化瘀解毒方、黄芪、莪术、重楼、壁虎、顺铂药物干预21 d后,取瘤组织,行免疫组化检测。结果:(1)成瘤体积比较:给药后瘤体积及差值比较,各组与空白组均有显著性差异(P<0.05);全方组分别与黄芪组、重楼组、壁虎组、空白对照组等比较有显著性差异(P<0.05)。(2)对HIF1a的影响:全方组与黄芪组比较无显著性差异(P>0.05),与其它组比较有显著性差异(P<0.05);各组与空白组比较有显著性差异(P<0.05);莪术组、壁虎组、顺铂组间比较无显著性差异(P>0.05)。(3)对E-cad影响:全方组与黄芪组比较无显著性差异(P>0.05),莪术、重楼、壁虎组与空白组比较无显著性差异(P>0.05)。结论:(1)益气化瘀解毒方全方较单味中药具有较强的抑瘤作用,其组方中药如莪术、重楼、壁虎均有一定程度抑瘤作用,莪术抑瘤作用较强;组方内主要单味中药发挥了协同抑瘤效应,使整方呈现出最强的抑瘤效应。(2)益气化瘀解毒方能抑制HIF1a表达,促进E-cad表达,可能通过阻断上皮间质转化而发挥抗肝癌转移侵袭作用,组方中药对其作用各有侧重,但作为整方的有机组成从不同环节发挥了协同作用。Objective:To observe the effect of Yiqi Huayu Jiedu (YQHYJD) formula and its separate on nude mice transplanta- tion tumor of hepatoma cells and the expression of HIF1 a and E-cad. Methods : Models of nude mice transplantation tumor are di- vided into 7 groups, treated by YQHYJD, astragalus, rhizoma zedoariae, paris polyphylla, gecko and cisplatin, after 21 days, immu- nohistochemical detection in tumor tissue. Results : ( 1 ) The effect on tumor volume : There was significant difference between the black and the other except Astragalus ( P 〉 0. 05 ). There was significant difference between cisplatin and the other( P 〈 0. 05 ). There was significant difference between YQHYJD and the separates (P 〈 0. 05 ) . (2) The effect on HIF1 a:There was significant difference between the black and the other( P 〈 0. 05 ). There was significant difference between YQHYJD and the other drugs ex- cept Astragalus ( P 〈 0. 05 ). ( 3 ) The effect on E-cad : There was significant difference between YQHYJD and the other drugs ex- cept Astragalus( P 〈 0. 05 ). There was no significant difference between rhizoma zedoariae, paris polyphylIa, gecko and the black ( P 〉 0. 05). Conclusion : Yiqi Huayu Jiedu formula plays a better effect of anti-tumor than Single herbs, rhizoma zedoariae, paris polyphylla, and gecko has the effect of anti-tumor, Yiqi Huayu Jiedu formula can inhibit the expression of HIF1 a and E-cad. rhizoma zedoariae, paris polyphylla, and gecko has on effect on E-cad.
