摘　　要：目的：建立益肺通络颗粒水提液总多糖的含量测定方法.方法：以葡萄糖为对照品,通过单因素试验,优选苯酚—硫酸法测定总多糖的最佳显色条件.结果：最佳显色条件为：加5％苯酚1.0mL,浓硫酸6.0mL,在100℃的条件下反应20min,冰水中冷却至室温.葡萄糖质量在16.6～124μg范围内与吸光度呈良好线性关系（r=0.999 1）,平均回收率为100.6％,RSD为2.5％.结论：该法快速准确、稳定方便,可用于益肺通络颗粒工艺研究中总多糖的含量测定.Objective： To establish the content determination method of total polysaccharides in aqueous extracts of Yifei Tongluo Granules. Methods. With grape sugar as reference substance,the best coloration conditions of phenol-sulfuric acid method for assa- ying total polysaccharides were optimized by single factor test. Results： The best coloration conditions were as follows., adding 1. 0mL of 5% phenol and 6.0mL of sulfuric acid,reacting at 100℃ for 20min, and then cooling to room temperature in ice water. The mass of glucose had a good linear relationship with absorbance at the range of 16.6 μg~ 124μg（r= 0.999 1）, and the average recovery was 100.6 %, RSD 2.5 %. Conclusion：The method is fast, accurate, stable and convenient, which can be available for determination for total polysaccharide in the preeess studies of Yifei Tongluo Granules.
