摘 要:目的:研究大孔吸附树脂纯化蒺藜呋甾皂苷、总黄酮的工艺条件。方法:以蒺藜呋甾皂苷、总黄酮含量及其转移率为指标,采用单因素试验法,优选树脂富集纯化蒺藜呋甾皂苷和总黄酮的最佳工艺。结果:纯化最优工艺条件:上样量为0.4g/mL,树脂径高比为1∶8,4BV水以2mL/min流速洗脱,分别用5BV50%乙醇、2BV/h 70%乙醇洗脱。D101树脂纯化后蒺藜呋甾皂苷纯度由22.59%提高到53.54%,增加了1.37倍;总黄酮的纯度由12.46%提高到32.11%,增加了1.58倍。结论:D101大孔吸附树脂可用于蒺藜呋甾皂苷和总黄酮的纯化。Objective : To discuss purification technology conditions of furostanol saponins and total flavonoids from Tribulus terres- tris L. by macroporous resin. Methods: With the the content of furostanol saponins and total flavonoids as indexes, purification technology conditions were optimized by single factor experiment. Results:Optimum purification conditions were as follow: concen- tration of sample liquid was 0.4g/ml, ratio of diameter to height of macroporous resin column was 1 : 8, them were eluted by 4BV water at 2mL/min, 5BV 50% and 70% ethanol at 2BV/h respectively. The purity of furostanol saponins reached 53.54%, which was two times higher than that before purification. And the purity of total flavonoids reached 32.11% ,which was two times higher than that before purification. Conclusion:D101 Macroporous Resin is suitable for the purification of furostanol saponins and total flavonoids from Tribulus terrestris L.
