摘 要:目的探讨短期锰暴露对大鼠睾丸内谷氨酸(Glu)、γ-氨基丁酸(GABA)、谷氨酰胺(Gln)含量的影响。方法将32只健康成年SPF级雄性SD大鼠随机分为4组,分别为对照(无菌生理盐水)组和7.5、15、30 mg/kg氯化锰染毒组,每组8只。采用腹腔注射方式进行染毒,每天1次,每周5次,连续染毒4周。测定大鼠睾丸组织内Glu、GABA、Gln含量。结果与对照组比较,30 mg/kg氯化锰染毒组大鼠睾丸内Gln、GABA含量及15、30 mg/kg氯化锰染毒组大鼠睾丸内Glu含量均较低,差异有统计学意义(P<0.05)。随着氯化锰染毒浓度的升高,大鼠睾丸内Glu、Gln、GABA的含量均呈先升高后下降的趋势。结论过量锰暴露可影响雄性小鼠性腺的GABA能系统,导致睾丸组织的Glu、Gln及GABA的含量降低。Objective To understand the effects of short-term exposure to manganese on amino acid neurotransmitters, including glutamate (Glu),glutamine (Gln),gamma-am nobutyric acid (GABA) in the testis of male rats. Methods Thirty-two SD male rats were randomly divided into the control,7.5,15,30 mg/kg body weight MnC12 groups. The rats in experiment groups received daily intraperitoneally injections of MnCl2 solution,while the rats in control group received daily intraperitoneally injections of normal saline (NS),once a day,five days per week,for four consecutive weeks. The concentrations of Glu, Gin and GABA in testis of rat were detected b,, the high performance liquid chromatography. Results After four-week exposure,compared with the control,the concentrations' of Gin and GABA in testis in 30 mg/kg MnClz-exposure group and the concentrations of Glu in testis of 15,30 mg/kg MnC12-exposure groups decreased (P〈0.05). Conclusion Excessive manganese exposure may affect the concentration of Glu, Gln and GABA in testis of adult rats.
