摘 要:目的:探讨血管内皮生长因子(vascular endothelial growth factor,VEGF)和miR-125b在新生儿缺氧缺血性脑病(hypoxic-ischemic encephalopathy,HIE)患儿和正常新生儿血液中表达水平及相关性。方法:收集20例正常新生儿和20例HIE患儿血液标本,分离血浆和外周血单个核细胞(peripheral blood mononuclear cell,PBMC),利用real-time PCR检测PBMC中miR-125b表达,ELISA方法检测血浆中VEGF含量。结果:与正常对照组相比,HIE患儿PBMC中miR-125b表达显著降低(P=0.00002),而血浆中VEGF水平显著升高(P=6.26×10–8);同时在正常组中,miR-125b表达与VEGF水平无相关性(r=0.016,P=0.946,n=20),但在HIE组(r=–0.505,P=0.023,n=20)二者却呈显著负相关。结论:新生儿脑缺血缺氧时可能通过降低miR-125b表达从而升高VEGF表达。Objective: To explore the expression attd correlation between vascular endothelial growth factor (VEGF) and miR-125b in healthy and hypoxic-ischemic encephalopathy (HIE) neonatus. Methods: A total of 20 controls and 20 HIE samples were recruited in this study, plasma and peripheral blood mononuclear cell (PBMC) were prepared for detecting the miR-125b expression and VEGF concentration using real-time PCR and ELISA kit, respectively. Result: Compared with controls, miR-12Sb expression in PBMC was significantly decreased (P=0.00002) but VEGF concentration in plasma was increased in HIE group (P=6.26×10-8). Meanwhile, there was no significant correlation between miR-125b expression and VEGF level in controls (r=0.016, P=0.946,n=20). However, significant correlation was observed in HIE group (r=-0.505, P=0.023, n=20). Conclusion: The VEGF level was increased possibly due to the down-regulation ofmiR-125b in HIE neonatus.
