血站型去白红细胞游离血红蛋白含量变化的研究

摘　　要：目的观察去白红细胞f-Hb含量及其变化。方法国产去白四联袋采血,离心分离后制备成悬浮红细胞并过滤为去白红细胞。未滤的留样对照。采血后第1 d、第7 d、第14 d、第21 d、第28 d、第35 d分别测f-Hb含量。结果去白红细胞f-Hb第1 d(71.04 mg/L±8.31 mg/L)高于悬浮红细胞(22.41 mg/L±4.3 mg/L)(P<0.05);14 d后与保存时间呈正相关;14 d与以后各时间段比,差异有统计学意义;第35 d(147.63 mg/L±15.43 mg/L),低于悬浮红细胞(152.74 mg/L±10.75 mg/L),但差异无统计学意义。结论过滤会影响f-Hb,但与本课题另一实验结果比较,离心和成分制备,对f-Hb的影响大于滤器去白过程,并决定f-Hb的总量。Objective To study the content and changes of free hemoglobin （ f - Hb） of leucocyte - poor red cells （LPRCs）. Methods The whole blood was collected by homemade leucocyte - removing quadruple bag to prepare the red blood cells sus- pension（CRCs） by centrifugal separation. Then the CRCs was filtrated to obtain the LPRCs, and the the unfiltered suspension was used as controL. The f-Hb contents of LPRCs and CRCs were determined on the 1st, 7th, 14th, 21st, 28th, 35th days. Results The content of f - Hb in LPRCs （71,04 mg/L ± 8.31 mg/L） was higher than CRCs （ 22.41 mg/L± 4.3rag/L） on the 1st day（P 〈0.05） , and kept stable in the first 14 d. However, it positively related with the preservation time from the 14th day, and the content of f- Hb was greatly different from that of the 21st, 28th, 35th days respectively（P 〈0.05）. The f- Hb content in LPRCs（ 147.63 mg/L ± 15.43 mg/L） was lower than CRCs（ 152.74 mg/L ± 10.75 rag/L） , but the difference had no statistical significance. Conclusion The content of f - Hb can be influenced by filtration. Compared with other relevant ex- perimental results, the centrifugal process and preparation process had greater influence on the test results than the filtration process, and also decide the total amount of f-Hb.
