摘  要:目的将荧光环介导等温扩增技术(loop-mediated isothermal amplification,LAMP)应用于饮食及公共场所服务业从业人员健康体检肛拭子标本中沙门菌的检测,评价其作为快速检测方法的可行性和可靠性。方法对荧光LAMP技术进行特异性、灵敏度试验,然后利用荧光LAMP技术对采集的1 536例肛拭子标本进行沙门菌检测,并以传统分离培养法进行平行对照实验。结果该方法仅对沙门菌产生特异性扩增,灵敏度可达2.98×102CFU/ml。经统计分析,荧光LAMP方法检出阳性率高于培养法(P<0.05),灵敏度100%、特异度99.41%、符合率为99.41%。结论荧光LAMP技术应用于沙门菌检验,其检测成本较低、特异性高、灵敏度高,可以在简易的实验条件下快速检测,适合基层检验部门对健康体检人群进行沙门菌的初步筛查工作。Objective To apply the fluorescence loop - mediated isothermal amplification (LAMP) to test anal swab samples of the physical examination among employees and evaluate its feasibility and reliability as a rapid detection method for Salmonella. Methods According to specificity and sensitivity test was carried out on the fluorescence LAMP method, and then using the fluorescence LAMP method assayed 1 346 cases of anal swab specimens, compared with the isolated culture method. Results With the fluorescence LAMP method, only Salmonella had specific amplification reactions, and the sens!tivity of fluo- rescence LAMP assay was 2.98 ~ 102 CFU/ml. By the statistical analysis, fluorescence LAMP method for the positive rate of detection is higher than culture method (P 〈 0.05), the sensitivity, specificity and coincidence rate were 100% ,99.41%, 99.41%, respectively. Conclusion The fluorescence LAMP assay had the advantages of high specificity and sensitivity, rapidi- ty and low cost,which can be used as a primary screen method to detect Salmonella at basic clinical laboratory.
