摘 要:目的研究DNA修复酶基因XRCC1、XRCC2、ERCC1、ERCC2和RAD52单核苷酸多态性在1,3-丁二烯(BD)致外周血淋巴细胞染色体损伤遗传易感性中的作用。方法选取187名BD作业工人和60名行政人员作为研究对象,采用胞质分裂阻滞微核试验(CBMN)评价个体外周血淋巴细胞染色体损伤水平,聚合酶链反应-限制性片段长度多态性方法分析XRCC1、XRCC2、ERCC1、ERCC2和RAD52基因多态性。结果在校正年龄、性别、吸烟、饮酒影响因素后,BD作业工人CBMN率为7.19‰±5.43‰显著高于对照组3.18‰±2.77‰,P<0.01;使用协方差分析,发现暴露组具有XRCC1 C26304T位点CT基因型个体CBMN率显著高于CC基因型个体(8.73‰±6.20‰,6.16‰±4.92‰,P<0.05);XRCC1 G28152A位点GG基因型个体CBMN率显著高于AA基因型个体(6.97‰±5.0‰,3.69‰±3.66‰,P<0.05);ERCC1 C19007T位点TT基因型个体CBMN率显著低于CC基因型个体(3.88‰±3.58‰,7.62‰±5.63‰,P<0.05)。结论XRCC1 C26304T、G28152A和ERCC1 C19007T位点的多态性可能影响BD作业工人染色体损伤的易感性。Objective To explore the relationship between chromosomal damage induced by 1,3-butadiene(BD) and polymorphisms of DNA repair gene of XRCC1,XRCC2,ERCC1,ERCC2 and RAD52.Methods 187 BD-exposed workers and 60 controls were recruited in this study.Chromosomal damage in peripheral blood lymphocytes was measured by cytokinesis-block micronucleus(CBMN) assay.And the polymorphisms of XRCC1,XRCC2,ERCC1,ERCC2 and RAD52 were detected by PCR-RFLP.Results By comparing the CBMN of the two groups we found that the frequency in exposed workers was higher than that in control group(P 〈 0.01).Multivariate analysis of covariance revealed that in BD-exposed workers,the XRCC1 CT genotype of C26304 T exhibited significantly higher CBMN frequency than that of CC genotypes(8.73‰ ± 6.20‰,6.16‰ ± 4.92‰,P 〈 0.05); The XRCC1 GG genotype of G28152 A exhibited higher CBMN frequency than that of AA genotypes(6.97‰ ± 5.0‰,3.69‰ ± 3.66‰,P 〈 0.05) significantly; The ERCC1 TT genotype of C19007 T exhibited lower CBMN frequency than that of CC genotypes significantly(3.88‰ ± 3.58‰,7.62‰ ± 5.63‰,P 〈 0.05).Conclusion The genetic polymorphisms of XRCC1 G28152 A and XRCC4 A245 G could influence the susceptibility to DNA damage in BDexposed workers.
