摘 要:为揭示促性腺激素抑制激素(GnIH)在公猪生殖发育中的作用,本研究以15头雄性小梅山猪为研究对象,用免疫组化和半定量RT-PCR方法研究3、30、60、90和120 d的公猪(n=3)GnIH阳性细胞在下丘脑-垂体-睾丸轴的分布定位及在下丘脑的发育学变化。结果表明:GnIH在下丘脑主要分布于室旁核,GnIH阳性神经元呈纺锤形和多角形,且在下丘脑室旁核GnIH阳性神经元数目随日龄增长而上下波动,形态趋向成熟;在垂体分布非常广泛,无论是腺垂体还是神经垂体均有强烈的免疫阳性反应;在睾丸集中分布于生精小管内部,可见强烈的免疫阳性反应,在间质细胞也可见免疫阳性反应。GnIH mRNA的丰度随日龄增长而呈规律性波动,在120 d出现显著降低(P<0.05)。结论:GnIH分布于小梅山公猪下丘脑-垂体-睾丸轴,并对其发育有调节作用。Gonadotropin-inhibitory hormone( GnlH), a key regulator of vertebrate reproduction, was recently identified in the Japanese quail. GnIH was reported to have many important physiological roles in reproduction, behavior,feeding, stress, etc. The potential role of GnIH in regulating reproductive axis was little reported, especially in domestic animals, and there was no research about reproductive regulation of GnIH to boars in literature. Thus, to investigate the potential role of GnIH in the hypothalamus-pituitary-testis(HPT) axis of boars, we systematically examined the expression and localization of GnIH in the HPT axis during the development of boarsand, and 15 boars of Xiaomeishan pigs(3,30,60,90,and 120 days after birth,n=3)were used in this study. We investigated the distribution of GnIH in the HPT axis of boars and developmental changes in the hypothalamus by immunohistochemistry and semiquantitative RT-PCR. The results showed that the GnIH immunoreactivities were found in the hypothalamus,pituitary and testis. In the hypothalamus, GnIH positive neurons in the paraventrieular nucleus (PVN), were seen in spindle and polygon. The number of the positive GnIH neurons in the paraventricttlar hypothalamic nucleus (PVN)show a fluctuating change and tend to be mature along with the age growth. In the pituitary, GnIH positive cells were localized in the adenohypophysis and neurohypophysis. In the testis, GnIH was mainly localized in the Sertoli cells, spermatogenic cells and leydig cells. The expression pattern of GnIH in the hypothalamus of boars during the development of boars were also systemically investigated. The hypothalamic level of GnIH mRNA was the highest on 3 d, followed by a significant decrease on 30 d and then increase on 60 d and finaly declined from day 60 d to 120 d,with the lowest expression level on 120 d (P〈0.05).
