摘  要:目的 评价右美托咪定对老龄大鼠术后海马脑源性神经营养因子(BDNF)及胆碱乙酰转移酶(ChAT)的影响.方法 老年雄性SD大鼠144只,18~20月龄,体重450~500 g,随机分为三组:正常对照组(C组,n=48),骨折复位模型组(M组,n=48)和右美托咪定组(D组,n=48).D组于骨折复位手术开始前20 min输注右美托咪定负荷剂量3.0 μg/kg,之后以3.0 μgkg-1·h-1持续输注至手术结束.术前1d、术后1、3、7d进行Morris水迷宫测试,记录逃避潜伏期和游泳总距离.各时点水迷宫实验结束后处死取海马组织,采用ELISA法检测海马BDNF和ChAT的含量,采用免疫组化法测定海马BDNF和ChAT的表达.结果 与C组比较,M组术后1、3d和D组术后1d水迷宫测试的逃避潜伏期及游泳总距离明显延长,海马BDNF和ChAT的表达明显降低(P<0.05).与M组比较,D组术后1、3d水迷宫测试的逃避潜伏期及游泳总距离明显缩短,而海马BD-NF和ChAT的表达明显增加(P<0.05).与术后1d比较,M组术后7d和D组术后3、7d水迷宫测试的逃避潜伏期及游泳总距离明显缩短,而海马BDNF和ChAT的表达明显增加(P<0.05).结论 右美托咪定可改善老龄大鼠术后认知功能,其机制可能与其促进海马BDNF及ChAT的释放有关.Objective To investigate the effects of dexmedetomidine on postoperative cognitive function in aged rats and hippocampal brain-derived neurotrophic factor (BDNF) and choline acetyltransferase (ChAT) expression.Methods One hundred and forty-four aged male SD rats,weighing 450-500g,were randomly divided into 3 groups,normal control group (group C,n=48),reduction of the fracture model group (group M,n=48),dexmedetomidine group (group D,n=48).In group D dexmedetomidine 3 μg/kg was injected iv immediately after,followed by infusion of dexmedetomidine at a rate of 3μg·kg-1 ·h-1 until the operation over.Morris water maze tests were performed on preoperative 1 day and postoperative 1 day,3 days and 7 days.The escape latency and swimming distance were recorded.After water maze test,the animals were sacrificed and their hippocampi were removed in each point for determination of BDNF and ChAT contents by ELISA,the expression of hippocampal BDNF and ChAT determination by immumohistochenistry.Results Compared with group C,the escape latency was prolonged and swimming distance of the water maze test was extended on postoperative 1 day,3 days in group M and postoperative 1 day in group D,the expression of BDNF and ChAT were decreased (P<0.05).Compared with group M,the escape latency and swimming distance of the water maze test were shortened on postoperative 1 day,3 days in group D,the expression of BDNF and ChAT were increased (P<0.05).Compared to postoperative 1 day,the the escape latency and swimming distance of the water maze test was shortened on postoperative 3 days in group D and postoperative 7 days in group M,and the expression of BDNF and ChAT were increased (P< 0.05).Conclusion Dexmedetomidine can improve cognitive function in aged rats,the mechanism may be related with dexmedetomidine enhances the expression of BDNF and increase the expression of ChAT in hippocampus.
