酸脂清胶囊对实验大鼠急性痛风性关节炎模型IL-1β、TNF-α的影响

摘　　要：目的通过观察酸脂清胶囊不同提取物对实验大鼠痛风性关节炎模型IL-1β和TNF-α的影响,研究酸脂清胶囊抗炎作用机制及对痛风性关节炎软骨破坏的作用,指导临床用药。方法酸脂清胶囊不同提取物(水提法,醇提法)大鼠灌胃7天后,尿酸钠关节腔注射造成大鼠急性痛风性关节炎模型,各组分别于造模后9h和12h随机抽取8只大鼠测量踝关节直径;下腔静脉取血,Elisa法检测炎症因子IL-1β和TNF-α。结果水提组、醇提组和秋水仙碱组致炎因子较模型组均有不同程度的降低,以水提组降低最为明显。结论酸脂清胶囊通过抑制IL-1β和TNF-α的水平,达到抗炎、保护关节软骨的作用,尤以水提组为优。Objective By observing the effective of the experimental gouty arthritis model＇s IL-1beta and TNF-alpha in rats to find its clear anti-inflammatory mechanism of action and its effective to cartilage destruction and advising clinical medicine.Methods With the extraction of proprietary Chinese medicines Suanzhiqing capsules（ water extraction,ethanol extraction） intragastric injection administration to the rats of acute gouty arthritis caused by uric acid sodium model for 7 days. And respectively at 9h and 12h randomly selected six rats in each group to measure the diameter of the ankle joint then take the inferior vena cava blood with ELISA detection to disclose the amount of inflammatory cytokines. Results Chinese medicine and western medicine（ colchicine） groups＇ proinflammatory cytokines were significantly lower compared with the model group,and the water extract group loss was the most obvious. Conclusion Suanzhiqing capsules worked by control the level of IL-1β and TNF-α to anti-inflammatory and protect articular cartilage. The best role was in the water extract group.
