温阳解郁不同组方对母婴分离/社会击败应激小鼠影响的比较研究

摘　　要：目的 用温阳解郁三种不同组方对模拟早年不良经历的母婴分离小鼠进行干预,比较其治疗效果并探讨其作用机理。方法 选用从出生起连续10天、每天3h母婴分离的12~20周成年C57BL/6J小鼠遭受社会击败应激塑造抑郁症模型。分别以逍遥散、二仙汤和温阳解郁复方进行6周的干预,通过旷场、高架十字迷宫及强迫游泳实验观察行为模式变化并运用原位杂交检测不同脑区BDNF的表达。结果 模型小鼠在旷场实验中央区域的活动减少,高架十字迷宫实验中开放臂和中央交汇区进入次数及停留时间均明显减少,强迫游泳不动时间明显延长,提示该模型已有明显的抑郁或焦虑表现。二仙汤和温阳解郁复方均能显著提高中央交汇区进入次数（P〈0.05）,温阳解郁复方能显著缩短强迫游泳不动时间（P〈0.05）。除前额皮质（PFC）外,模型小鼠各脑区的BDNF表达均明显下降,各治疗组均能明显提高各脑区BDNF mRNA的表达量。结论 温阳解郁复方对抑郁症有明显治疗作用,可能是温阳和解郁互相配合的结果。Objective To study the mechanisms of warming yang and soothing liver compound（ XSF） treating maternal separation mice,and the basis for the treatment of depression. Methods The adult C57 BL/6J mice which experienced 10 d maternal separation after birth（ MS） were treated with Erxian,Xiaoyao and XSF formulas for 6 weeks,3 hours per day,then their behavior changes were observed by open field,elevated plus maze（ EPM） and forced swimming test（ FST）,and BDNF mRNA was detected by in situ hybridization. Results Erxian formula and XSF markedly increased Central Zone Entries of MS mice in EPM（ P 0. 05）,XSF significantly shortened the immobile time of mice in FST（ P 0. 05）,and all the three formulas obviously enhanced the expression of BDNF mRNA in different brain areas. Conclusion Warming yang and soothing liver formula has significant improvement effects on mice with depression,coordination of warming yang and smoothing the liver may be the main mechanism.
