HPLC测延胡索中延胡索乙素和原阿片碱的含量

摘　　要：目的：采用高效液相色谱法同时测定延胡索药材中延胡索乙素和原阿片碱的含量.方法：色谱柱：kromasil-C18柱,（5μm,250mm×4.6mm）,流动相：乙腈：0.1％磷酸水溶液（三乙胺调pH值为6）（19：81）,检测波长：281nm,检测器：Waters 2487紫外检测器.结果：延胡索乙素在3.96～33μg/ml范围内线性关系良好;原阿片碱在3.0～50μg/ml范围内线性关系良好.结论：本含量测定方法操作简单,灵敏度高,准确性好,重现性好,可用于测定延胡索生物碱的含量.Objective：The research aimed to establish a method for determination of dl-Tetrahydropalmatine and protopine in Corydalis yanhusuo. Methods：HPLC separation was carried on a column of kromasil-Cl8 （51xm,250mmX4.6 mm）, with the mobile phase of 0.1% phosphate bufferacetonitrile （19：81）at the flow rate of 1.0ml/min by UV-detcctor, and the detection wavelength was set at281 nm, Results：d1-Tetrahydropalmafine showed a good linearity within the range of 3.96-33μg/ml, protopine showed a good linearity within the range of 3.0-50μg/ml. Conclusion： The method was simple, accurate and reproducible. It could be used for the quality control of Corydalis yanhusuo.
