摘  要:目的:对比观察电针内关穴对心肌缺血大鼠L型钙离子通道β2亚基基因表达的影响。方法:选取健康雄性SPF级大鼠50只,随机分为空白组、模型组、内关组、列缺组和非经穴组。除空白组外,以异丙肾上腺素诱导建立心肌缺血大鼠模型,电针治疗选用疏密波,频率2/50Hz,强度2-4mA,以大鼠肢体微微颤动为宜,留针20min,每日1次,连续7d。记录各组大鼠心电图Ⅱ导联T波振幅的变化,检测各组大鼠血清心肌肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)的含量及L型钙离子通道β2亚基基因表达。结果:与空白组比较,模型组心电图T波振幅显著降低(P<0.01),CK-MB、LDH及L型钙离子通道β2亚基基因表达均显著提高(P<0.01,P<0.05);与模型组比较,内关组大鼠心电图T波振幅显著提高(P<0.01),CK-MB和LDH表达水平显著降低(P<0.01),L型钙离子通道β2亚基mRNA表达受到显著抑制(P<0.01),列缺组与非经穴组也有所降低,但没有内关组明显(P<0.01)。结论:内关穴对心肌细胞钙离子跨膜通道表达有特异性调控作用。Objective: To compare the effects of Neiguan (PC6) on the gene expression of β 2 subunit of L-type voltage- gated Ca2+ channels in rats with myocardial ischemia. Methods: 50 male SPF rats were randomly divided into blank control group, model control group, Neiguan (PC6) group, Lieque (LU7) group, and non-acupoints group. All rats were injected isoproterenol (ISO) to establish myocardial ischemia rat model, except for the blank control group. All rats except for the blank control group were received electro-acupuncture treatment which choose condensation-rarefaction wave at frequencie 2/50Hz, with current of 2-4mA, at 20min retaining needle time, daily 1 time, last for 7 days. The amplitude values of T wave in rats ECG were recorded. The changes in the CK-MB and LDH levels and the gene expression of β 2 subunit of L-type voltage-gated Ca2+ channels were also determined in all groups. Results: The CK-MB and LDH levels and the gene expression of β 2 subunit of L-type voltage- gated Ca2+ channels in model control group and all electro-acupuncture treatment groups were significantly higher than those in the blank control group (P〈0.01, P〈0.05). The amplitude values of T wave in the Neiguan (PC6) group were significantly higher than those in the model control group (P〈0.01), while the LDH and CK-MB levels and the gene expression β 2 subunit of L-type voltage-gated Ca2+ channels in the Neiguan (PC6) group significantly lower (P〈 0.01). The LDH and CK-MB levels and the gene expression β2 subunit of L-type voltage-gated Ca2+ channels in the Lieque (LU7) and non-acupoints groups were also reduced, but the changes were not as obvious as those in the Neiguan (PC6) group (P〈0.01). Conclusion: Neiguan (PC6) can regulate specifically membrane-spanning calcium ion channels in myocardial cells.
