化痰祛湿活血方对非酒精性脂肪性肝炎大鼠脂联素及其受体的干预研究

摘　　要：目的:探讨脂联素(ADPN)及其脂联素受体2(AdipoR2)对非酒精性脂肪性肝炎(NASH)病理影响及化痰祛湿活血方干预作用。方法:采用高脂饲料联合四环素腹腔注射建立大鼠NASH模型。造模第2周采用化痰祛湿活血方高、中、低剂量和多烯磷脂酰胆碱(易善复)胶囊干预观察高脂模型。实验结束时,进行肝组织HE染色,ADPN、AdipoR2免疫组化染色,观察病理改变及ADPN、AdipoR2表达。结果:与对照组比较,化痰祛湿活血方高剂量能明显升高ADPN、AdipoR2蛋白表达(P<0.01),减轻肝脂肪变性及炎症程度。结论:化痰祛湿活血方可能通过上调ADPN、AdipoR2表达,减轻肝脏炎症,改善肝细胞脂肪沉积。Objective： To study the pathological effect of adiponectin and adiponectin receptors on nonalcoholic steatohepatitis （NASH）, and the intervention effect of Huatan Qushi Huoxue Recipe. Methods： The method of injecting oxytetracycline intraperitoneally and taking fat-rich food was used to establish NAFL model in rats. Two weeks after modeling, the treatment groups were treated with different doses （high, medium, low） Huatan Qushi Huoxue Recipe respectively while the control group was treated with Polyene Phosphatidylcholine （Yishanfu） Capsule. When the experiment finished, the pathologic slices of liver were observed by HE staining, and the expression of ADPN and AdipoR2 were detected by immunohistochemical staining. Results： Compared with Yishanfu group, the expression of ADPN, AdipoR2 protein were significant increased in high dose Huatan Qushi Huoxue Recipe group （P〈0.01）. It could reduce the degree of liver steatosis and inflammation. Conclusion： Huatan Qushi Huoxue Recipe could effectively reduce liver inflammation and improve fat deposition in the hepatocytes by up- regulating the expression of ADPN and AdipoR2.
