摘  要:目的:研究猪苓利水渗湿功效对膀胱癌大鼠模型水通道蛋白1(AQP1)、水通道蛋白3(AQP3)调节作用。方法:制作膀胱癌大鼠模型,分为生理盐水组、模型组、猪苓50mg/kg组、猪苓250mg/kg组、猪苓500mg/kg组,按组别分别药物干预后,以免疫组化技术和RT-PCR技术检测膀胱组织AQP1和AQP3表达情况。结果:膀胱癌大鼠模型诱导成功,猪苓干预膀胱癌大鼠模型后,对BBN诱导的膀胱癌模型有显著的抑制作用。与模型组比较,猪苓能明显降低膀胱组织AQP1和AQP3蛋白表达,猪苓250mg/kg组和猪苓500mg/kg组均可显著降低AQP1和AQP3 mRNA相对表达含量(P<0.05,P<0.01)。结论:猪苓利水渗湿功效可通过抑制AQP1和AQP3表达抑制膀胱癌发生发展。Objective: To study the diuretic and excreting dampness effect of grifola umbellate on the regulation of AQP1, AQP3 in rats with bladder cancer. Methods: The rat model of bladder cancer was established, and the rats were divided into NS group, model group, grifola umbellate groups (50mg/kg, 250mg/kg, 500mg/kg dose). After corresponding drugs intervention, the expression of AQP1 and AQP3 were measured by immunohistochemical method and RT-PCR. Results: The rat model of bladder cancer was induced successfully. After grifola umbellate intervened the rat model of bladder cancer, BBN-induced bladder cancer model had significant inhibited. Compared with the model group, the expression level of AQPI and AQP3 protein in bladder tissue decreased obviously, the relative expression of AQP1 and AQP3 mRNA decreased in the 250mg/kg and 500mg/kg dose of grifola umbellate (P〈0.05, P〈0.01). Conclusion: The diuretic and excreting dampness of grifola umbellate can inhibit the expression of AQP1 and AQP3 in bladder in order to inhibit the occurrence of bladder cancer.
