摘  要:对青岛百合(Lilium tsingtauense Gilg)复合种群6个局部种群的遗传多样性进行了RAPD分析.通过11个引物扩增到117个显性位点,其中多态性位点平均为62.86%,各百合群体多态性位点多少顺序为Y5、Y1、Y4、Y6、Y2、Y3.把RAPD多样性指标与样地的基本状况进行相关分析,发现海拔高度、样地坡度和植被类型与各个多样性指标呈正相关,样地坡向与各个多样性指标呈负相关.Nei's基因多样度指数(h)和Shannon信息指数(I)的统计结果表明,种群内h和I的平均值分别为0.203 4和0.309 0,这两个指数均为Y5最高,其次为Y1,Y3最低.不同位点在种群间的分化程度差别很大,基因分化度的波动范围为0.016 5~0.942 7,平均为0.378 3.种群间平均基因流为0.821 6,说明有遗传漂变的存在.6个百合种群间遗传一致度与遗传距离的无偏估计表明,最小的遗传距离在Y4和Y6之间,最大的遗传距离在Y3和Y5之间,遗传一致度则相反.种群聚类分析表明,Y1和Y2比较近;Y4和Y6比较近又和Y5联系在一起.Y3比较特殊,单独为一类.Genetic diversity of six Lilium tsingtauense metapopulations were studied with the method of RAPD.The genetic differentiation among populations was analyzed by STATISTICA.117 loci were amplified by RAPD.The means of genetic diversity within population was 62.86%.The order was population 5> population 1> population 4> population 6> population 2> population 3.Altitude,plot slope,vegetation types and various diversity index was positively correlated.But plots slope and various diversity index was negatively correlated.The result of Nei's gene diversity index (h) and Shannon (I) Information showed that the mean was 0.203 4 and 0.309 0.These two indices were both the highest in population 5,but the lowest in population 3.Genetic divergence of all RAPD loci among populations was different from 0.016 5 to 0.942 7,with the mean of 0.378 3.The change of gene flow was inverse to the change of genetic divergence.The means of gene flow was 0.821 6,less than 1,indicating that there was genetic drift among populations.From Nei's unbiased measures of genetic identity and genetic distance by RAPD,it was conducted that the lowest genetic distance was between population 4 and population 6,with the highest genetic identity between them.
