摘 要:采用热处理法从海南省佳西热带雨林土壤中分离到147株芽胞杆菌,并利用16S rDNA PCR -RFLP与序列分析技术对其遗传多样性进行了研究。16S rDNA PCR -RFLP 酶切图谱 UPGMA 聚类分析结果表明,在100%的相似性水平上,这些芽胞杆菌分属13个遗传类群。不同遗传类型代表菌株的16S rRNA 基因序列分析结果显示,它们分布在 Bacillaceae、Planococcaceae 和 Paenibacillaceae 科的 Bacillus、Lysinibacillus、Paucisaliba-cillus、Bhargavaea 和 Paenibacillus 五个属,其中 Bacillus 为优势属(占50%);有3株芽胞杆菌的16S rRNA 基因序列与数据库中相应模式菌株的最大相似性在98.3%~98.9%之间。结果表明,佳西热带雨林土壤中芽胞杆菌有着较为丰富的遗传多样性。147 Bacillus-like strains were isolated from Jiaxi tropical rain forest soil of Hainan using heat treatment method. The genetic diversity research was carried out with 16S rDNA PCR-RFLP and sequence analysis. At the level of 100% similarity,cluster analysis results of UPGMA of 16S rDNA PCR-RFLP enzyme digestion atlas showed that these Bacillus isolates belonged to 13 genetic groups. 16S rRNA gene sequence analysis of different genetic type of representative strains showed that they were distributed in five genera:Bacillus,Lysinibacillus,Paucisalibacillus, Bhargavaea and Paenibacillus of the families of Bacillaceae,Planococcaceae and Paenibacillaceae,and Bacillus was the dominant one(accounted for 50% ). Based on the comparison of 16S rRNA gene sequences,the high levels of 16S rRNA gene similarities among them three strains with the corresponding type strains in the data base ranged from 98. 3% ~ 98. 9% . The results showed that cultivable Bacillus-like species from tropical rain forest soil possessed fairly rich genetic diversity.
