摘  要:目的研究冷、热两种工艺加工的鹿茸提取物,对正常、慢性再生障碍性贫血患者骨髓的单个核细胞(mononuclear cell,MNC)体外增殖、分化的影响,为临床合理应用提供实验依据。方法从10例健康者、10例慢性再生障碍性贫血(aplastic anemia,AA)患者骨髓中获取MNC,分别在普通培养液(阴性对照组),加热炸茸C组分的培养液(试验组1)及加冷工艺茸A、B、D、E组分的培养液(试验组2)中培养。MTT比色法检测细胞的增殖活性;CFU-mix集落半固体培养观察集落生长情况;细胞免疫化学染色测定bcl-2、突变型p53的表达。数据处理采用单因素方差分析(one-way ANOVA),结果用x珋±s表示。结果正常骨髓组:试验组2细胞增殖活性及CFU-mix集落数均明显高于试验组1及阴性对照组;bcl-2和突变型p53的表达各组之间均无明显差异。AA骨髓组:试验组2中D、E组分细胞增殖活性和CFU-mix集落数均明显高于阴性对照组及试验组1。Bcl-2的表达:试验组2中D、E组分组显著高于阴性对照组和试验组1。突变型p53的表达三组之间均无明显差异。结论两种不同工艺的鹿茸相比,冷工艺茸具有更强的生物学效应,故在鹿茸的加工上推荐冷工艺。Objective To study the effects of pilose antler compositions, which extracted by two different technologies, on human bone marrow mononuclear cells from healthy persons and aplastic anemia patients. To guide theimprovement of processing technology of pilose antler and their future clinical use by testing the proliferation effect on the cultured cell in vitro. Methods Mononuclear cells (MNC) isolated from 10 healthy donors, 10 aplastic anemia patients were cultured in vitro. Each was divided into 3 groups 1. negative con- trol, with no pilose antler in medium ;2. experimental groups 1, with composition C from pilose antler of sika deer by heat-technology;3, experimental groups 2, with composition A, B, D, E from pilose antler of sika deer or red deer, by cool-technology. Proliferation of culture ceils were tested by methyl thiazolyltetrazolium (MTT) assay and CFU-mix colony forming test. Expression rate of mutant p53 and bcl-2 were tested by immunocytochemical dyeing. Data were analyzed by one-way ANOVA, SPSS 11.5. Results The group of normal bone marrow: ( 1 ) OD value and CFU-mix number: A, B, D, E groups are higher than experimental groups 1 and negative control. (2) The expression-rate of bcl-2 and mutant p53 has no difference among three groups. The group of AA bone marrow: ( 1 ) OD value and CFU-mix number: D, E groups are higher than experimental groups 1 and negative control. There is no difference between experimental groups I and negative control. (2) The expression-rate of bcl-2 : D, E groups are higher than negative control and experimental groups 1. There is no difference between experimental groups 1 and negative control. The expressionrate of mutant p53 is not different among three groups. Conclusions The compositions derived from cooltechnology have better effects on human bone marrow hematopoietic cells than that from heat-technology.
