摘  要:通过VY/4平板法、纤维蛋白平板法及纤维蛋白试管法,从兔粪、土、朽木及水等20个样品中筛选出5株可以产生抗凝溶栓双活性蛋白(1)的细菌。重点研究了菌株LDS33,发现其胞外产物有较强的抗凝溶栓活性,并初步分离纯化了产生的1。通过形态学、理化性质及16S rDNA序列的进化树分析确定菌株LDS33属于短小芽孢杆菌。优化条件,可知该菌株产1的最佳发酵条件为培养温度26℃、发酵培养基消毒前pH值为8.5、培养时间8 d。Five strains were screened that could produce the protein with anticoagulant and thrombolytic activities (1) by VY/4 plate, fibrin plate and fibrin tube from 20 samples such as rabbit manure, soil, deadwood, water, etc. A strain named LDS33 among them was studied. The experiment showed that its extracellular product had strong anticoagulant and thrombolytic activity. Product 1 were purified preliminarily from the strain LDS33. According to the morphology, physical and chemical properties and phylogenetic tree of 16S rDNA sequencing, the strains LDS33 was identified as Bacillus pumilus. The culture conditions for production of 1 by LDS33 were investigated and the results showed that the optimized fermentation conditions were temperature of 26 ℃, fermentation culture medium of pH 8.5 before disinfection and incubation for 8 d.
