摘 要:采用体外抗氧化活性(包括DPPH自由基清除法、ABTS自由基清除法和FRAP法)为导向,筛选红花抗氧化活性部位,随后对活性部位进行成分分离与抗氧化效应评价。从红花活性部位水部位中分离得到5个成分,分别鉴定为6-羟基山柰酚-3,6,7-三-O-β-D-葡萄糖苷(1)、6-羟基山柰酚-3-O-β-芸香糖苷-6-O-β-D-葡萄糖苷(2)、6-羟基山柰酚-3-O-β-D-葡萄糖苷(3)、羟基红花黄色素A(4)和脱水红花黄色素B(5)。通过对红花不同极性部位和单体成分进行抗氧化效应比较,发现水部位的抗氧化活性较显著,从中分离得到6-羟基山柰酚糖苷类和醌式查尔酮碳苷类主要活性物质。Bio-active components from Carthamus tinctorius were separated on the basis of antioxidant capacities in vitro. The antioxidant capacity was investigated on the basis of the ability to scavenge DPPH radical, ABTS radical and reduce Fe3 + of different polar fractions. Furthermore, the chemical compounds were isolated from bio-active fraction, and were evaluated for the antioxidative effects. Five major components were isolated and identified from water extract as 6-hydroxykaempferol 3,6,7-tri-O-β-D-glucoside( 1 ), 6-hydroxykaempferol 3-O-β-rutinoside-6-O-β-D-glucoside ( 2 ), 6-hydroxykaempferol 3-O-β-D-glucoside (3), hydroxysafflor yellow A (4) and anhydrosafflor yellow B($). By evaluating and comparing the antioxidative effects ofdifferent fractions and obtained compounds, the results showed that water extract displayed significantly high antioxidative activities and 6-hydroxykaempferol glycosides and quinochalcone C-glycosides were found as main contribution for antioxidant property.
