摘  要:马铃薯帚顶病毒(Potato mop-top virus,PMTV)是马铃薯上的重要病毒之一。本研究利用逆转录环介导等温扩增技术,建立了PMTV的RT-LAMP检测方法。根据PMTV高度保守的TGB序列设计了3组引物,进行引物筛选和条件优化。建立的RT-LAMP检测方法特异性强、灵敏度高,比PCR灵敏度高100倍,并可通过肉眼观察浊度或颜色反应进行结果判断。本研究为PMTV的检测提供了快速、灵敏的检测方法。Potato mop-top virus (PMTV) is one of the most important pathogens of potatoes. In this study, a reverse transcription loop-mediated isothermal amplification (RT-LAMP) method was developed for the detection of PMTV. Three primer sets were designed based on the highly conserved region of the TGB sequence of PMTV. After the optimization of primers and reaction conditions, the RT-LAMP method was specific and sensitive in detecting PMTV, the reaction was 100 times more sensitive than PCR. The detection result can be judged by visual observation of turbidity Or color reaction without complex equipments. This study will provide a rapid and sensitive diagnostic method to distinguish PMTV in potato.
