消痛膏中乌头碱、次乌头碱和新乌头碱的HPLC法测定

摘　　要：建立了高效液相色谱法测定消痛膏中的乌头碱、次乌头碱和新乌头碱。使用Hedera ODS-2柱,以乙腈∶四氢呋喃（25∶15）为流动相A,以0.1 mol/L乙酸铵溶液（每1 L加冰乙酸0.5 ml）为流动相B,梯度洗脱,检测波长235 nm。乌头碱、次乌头碱和新乌头碱分别在0.07～0.42ug、0.15～0.88ug和0.05～0.32ug范围内线性关系良好,回收率分别为92.7%、92.1%和93.0%,RSD分别为3.37%、2.09%和3.14%。An HPLC method was established for the determination of aconitine, hypaconitine and mesaconitine in Xiaotong patches. A Hedera ODS-2 column was used, with the mobile phase A of acetonitrile ： tetrahydrofuran （25 ： 15） and the mobile phase B of 0.1 mol/L ammonium acetate solution （adding 0.5 ml of glacial acetic acid per liter） by gradient elution, at the detection wavelength of 235 nm. The calibration curves were linear in the ranges of 0.07 - 0.42 μg, 0.15 - 0.88 μg and 0.05 - 0.32 μg for aconitine, hypaconitine and mesaconitine. Their average recoveries were 92.7%, 92.1% and 93.0%, with RSDs of 3.37%, 2.09% and 3.14%.
