摘  要:目的采用HPLC法测定川麦冬中的麦冬皂苷D,并比较其块根和须根中麦冬皂苷D的含量差异。方法采用Alltima C18色谱柱(250 mm×4.6 mm,5μm),乙腈-水(52∶48)为流动相,流速1.0 mL·min^-1,柱温30℃,进样量20μL,漂移管温度65℃,气体流速2.5 mL·min^-1。结果麦冬皂苷D 0.416~10.40μg与峰面积呈良好的线性关系(r=0.9993);平均回收率为99.12%,RSD=1.39%(n=9);麦冬块根和须根中麦冬皂苷D的平均含量分别为0.348‰和0.403‰。结论所用方法简便、快速、准确,适用于麦冬皂苷D的含量测定;麦冬须根和块根中麦冬皂苷D的含量无统计学差异。OBJECTIVE To determine and compare ophiopogonin D in the tuber root and fibrous root from Ophiopogon japonicus by HPLC. METHODS The separation was performed on Alltima C_18 (250 mm ×4. 6 mm ,5 μm)column with the mobile phase consisting of acetonitrile -water(52 : 48 )at the flow rate of 1.0 mL·min^-1. The column temperature was 30 ℃ and injection volume was 20μL. The drift tube temperature of ELSD was set at 65 ~C with the flow rate of 2. 5 mL·min^-1. RESULTS The linear range of ophiopogonin D was 0.416 - 10.40 μg( r = 0. 9993 ) , and the average recovery was 99.12% ( n = 9 ) with the RSD of 1.39%. The contents of ophiopogonin D in the tuber root and fibrous root from Ophiopogon japonicus were 0. 348‰ and 0. 403‰. CONCLUSION The method is simple, reliable and accurate, and can be used for the content determination of ophiopogonin D from Ophiopogon japonicas. The contents of ophiopogonin D in the tuber root and fibrous root of Ophiopogon japonicus are no different.
