摘  要:目的 研究赖诺普利对糖尿病大鼠坐骨神经结构蛋白的影响及对早期周围神经病变的保护作用.方法 25只健康雄性Wistar大鼠,链脲佐菌素(STZ)尾静脉注射,制备糖尿病大鼠模型,24只成功模型按随机数字表法分为糖尿病组(12只)和赖诺普利治疗组(12只),另选择10只健康大鼠作为正常对照.8周后,观察各组大鼠坐骨神经传导速度及超微结构的改变.采用免疫组化法测定坐骨神经神经丝蛋白(NFP)、髓鞘碱性蛋白(MBP)表达的变化.结果 糖尿病大鼠出现坐骨神经传导速度减慢(P<0.01)、痛阈减低(P<0.05)和结构异常,神经结构蛋白表达减少(P<0.01);给予赖诺普利治疗可提高坐骨神经传导速度(P<0.01),提高痛阈(P<0.05),减轻坐骨神经轴突和髓鞘萎缩、变性;增加坐骨神经NFP、MBP表达(P<0.01).结论 赖诺普利可能通过增加神经结构蛋白的表达发挥对早期糖尿病周围神经病变的保护作用.Objective To investigate influences of lisinopril on expression of sciatic nerve structural proteins and its protective effects on experimental diabetic peripheral neuropathy in the earlier stage.Methods Twenty-five healthy male Wistar rats with streptozotocin tail vein injection were prepared for diabetic rat models.They were randomly divided into diabetic group and lisinopril treatment group [20 mg/(kg · d),8 weeks].Ten healthy rats were used as normal controls.Sciatic nerve conduction velocity,pain threshold,and ultrastructure of rats were observed after 8 weeks.We determined the expression of neurofilament protein (NFP) and myelin basic protein (MBP) of sciatic nerve with immunohistochemistry in order to explore mechanisms of lisinopril.Results Diabetic rats appeared that sciatic nerve conduction velocity slowed down (P <0.01),the pain threshold reduced(P <0.05),and structural abnormalities.Lisinopril improved nerve conduction velocity (P < 0.01)and pain threshold (P < 0.05),extenuated atrophy and degeneration of axon and myelin in sciatic nerve,and increased the expression of NFP and MBP of sciatic nerve.Conclusions Lisinopril may exert neuroprotective effects through improving the expression of sciatic nerve structural proteins.
