摘 要:为研究高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)对断奶仔猪骨髓的损伤,本研究采用HP-PRRSV HuN4株接种30日龄的PRRSV抗原及抗体阴性的健康断奶仔猪,并在病毒感染后10 d检测仔猪骨髓内病毒感染及细胞凋亡情况。PCR和间接免疫荧光试验结果显示HuN4株感染仔猪骨髓内存在病毒;流式细胞计数分析和TUNEL法检测结果显示病毒感染仔猪的骨髓内出现大量的凋亡细胞。本实验结果表明,HP-PRRSV能够感染仔猪骨髓,引起仔猪骨髓损伤并导致机体免疫抑制。本研究为进一步探索HP-PRRSV感染诱导仔猪中枢免疫器官的损伤机制奠定了基础。To study the lesions in bone marrow of piglets infected with the highly pathogenic porcine reproductive and respiratory syndrome virus (HP-PRRSV), piglets were inoculated with HuN4 strain, and the virus and apoptotic cells in bone marrow were investigated. The results showed that PRRSV was detected in bone marrow of piglets infected with HP-PRRSV by PCR and IFA, and a large number of apoptotic cells were observed in bone marrow of these piglets by flow cytometry analysis and TUNEL assay. These results demonstrated HP-PRRSV had a broader range of tissue tropism compared to classical PRRSV strains, which was able to induce the depletion of bone marrow cells, and resulted in immunosuppression. These results provided a basis for studying the lesions mechanism on the center immune organs of piglets infected with HP-PRRSV.
