摘 要:为调查东北三省地区奶牛源葡萄球菌耐药现状、耐药产生的分子机制和菌株流行情况,本研究对东北三省4个不同奶牛厂分离的107株不同种属葡萄球菌进行耐药表型测定,并采用PCR方法检测分离株中携带的相关耐药基因;通过脉冲凝胶电泳(PFGE)对分离株的亲缘关系进行分析,同时对金黄色葡萄球菌进行多位点序列分型(MLST)。研究结果表明,同一奶牛场葡萄球菌克隆传播现象严重,PFGE分析后仅获得8株不同克隆的葡萄球菌。8株分离菌株对氨苄西林、克林霉素、红霉素、卡那霉素、和环丙沙星的耐药率分别为87.5%、87.5%、62.5%、50%和25%,对其余受试的6种药物(苯唑西林、四环素、氟苯尼考、利福平、万古霉素和利奈唑胺)均敏感。耐药基因检测结果显示,耐药基因blaZ/femB,lnuA/lnuB,erm(B),accA-aphD和gyrA基因Ser84(TCA)-I1e(TTA)的突变及grlA基因Ser80(CTC)-Phe(CTT)的突变是造成葡萄球菌对氨苄西林、克林霉素、红霉素、卡那霉素和环丙沙星耐药的主要原因。To investigate the prevalence and molecular mechanism of drug resistance in Staphylococcus spp. isolated from four dairy farms in part of Northeast China, a total of 107 Staphylococcus spp. isolates were collected and tested for the resistance-phenotype and-genotype by broth microdilution and PCR method, respectively. Pulsed-Filed Gel Electrophoresis (PFGE) and Multilocus Sequence Typing (MLST) were used for typing these isolates. PFGE analysis revealed that 8 isolates were classified to distinct PFGE types among the 107 Staphylococcus spp., suggesting the existence of prevalence clone in each farm. The resistance rates of the 8 isolates to ampicillin, lincomycin, erythromycin, kanamycin and ciprofloxacin were 87.5%, 87.5%, 62.5%, 50% and 25%, while all isolates were susceptible to oxacillin, tetracycline, florfenicol, rifampicin, vancomycin and linezolid, respectively. In addition, the blaZ/femB, lnuA/lnuB, erm (B) and aecA-aphD were detected in these resistant isolates. Furthermore, the mutations in gyrA gene (Ser-84-to-Ile) and in grlA gene (Ser-80-to-Phe) were found to be the main reason for resistance to ciprofloxacin in the 8 isolates.
