摘 要:研究旨在分析家鸭IFIT-5基因外显子2的多态性,以期为进一步研究家鸭IFIT-5基因与疾病抗性的相关性提供理论依据。根据GenBank上收录的IFIT-5基因序列设计特异性引物,采用PCR-SSCP方法对苏牧麻鸭、金定鸭、樱桃谷鸭与白羽番鸭这4个家鸭群体IFIT-5基因外显子2进行多态性分析。结果表明:在IFIT-5基因外显子2上检测到2种等位基因、2种基因型,存在1个SNP位点;在4个群体中均未检测到BB基因型个体,4个群体均不处于哈代-温伯格平衡,苏牧麻鸭跟金定鸭不同基因型分布差异不显著(P>0.05),而其余群体彼此不同基因型分布存在极显著性差异(P<0.01)。提示4个家鸭群体IFIT-5基因外显子2存在遗传多态性,不同家鸭群体在遗传基础上存在着一定的差异。This experiment was conducted to study the polymorphism of IFIT-5 gene exon 2 in duck aimed to provide a theoretical foundation for further research on correlation between duck IFIT-5 gene and disease resistance. The second exon of IFIT-5 gene in 4 duck populations including Cherry Valley duck,Jinding duck,White Muscovy duck and Sumu duck were detected by PCR-SSCP method with specific primer designed according to IFIT-5 sequence from GenBank. The results showed that 2 genotypes and 2 alleles were found in exon 2 of IFIT-5 gene which contained 1 SNP. BB genotype was not found in 4 duck populations. The distribution of these genotypes in all populations was not consistent with Hardy-Weinberg equilibrium. There were significant differences (P〈0.01)among different populations except Sumu duck and Jinding duck (P〉0.05). These results indicated that genetic diversity of IFIT-5 gene of 4 duck populations was detected in exton 2.
