摘  要:目的:考察罗仙子提取物对单核细胞与血管内皮细胞黏附的影响。方法:制备相对分子质量30 000以下罗仙子提取物;考察罗仙子提取物对血管内皮细胞EA.hy926活力的影响;肿瘤坏死因子(TNF-α)诱导建立单核细胞THP-1和EA.hy926黏附模型,采用虎红染色,考察不同浓度罗仙子提取物对EA.hy926与THP-1黏附性的影响;采用ELISA考察罗仙子提取物对TNF-α诱导的EA.hy926分泌单核细胞趋化蛋白-1(MCP-1)和血管细胞黏附分子-1(VCAM-1)的影响。结果:罗仙子提取物(浓度≤4×102μg/mL时)对EA.hy926未见明显的增殖或抑制作用(P>0.05);罗仙子提取物(浓度≥6.25μg/mL时)可浓度依赖性降低两细胞之间的黏附作用;罗仙子提取物各浓度均能显著地降低TNF-α诱导的MCP-1表达(P<0.01),罗仙子提取物(浓度≥50μg/mL时)可显著地降低TNF-α诱导的VCAM-1的表达(P<0.01)。结论:罗仙子提取物可通过抑制TNF-α诱导的内皮细胞分泌MCP-1和VCAM-1,阻滞单核细胞与血管内皮细胞之间的黏附作用,从而发挥抗动脉粥样硬化作用。AIM: To investigate the effects of luoxianzi extractive on the adherence rate of monocyte and vascular endothelial cell. METH- ODS. The luoxianzi extractive which molecular weight was less than 30 000 was prepared; the influence of luoxianzi extract on vascular endo- thelial cells EA. hy926 s vitality was investiga- ted the THP-1 and EA. hy926 adhesion model induced by tumor necrosis factor (TNF-α) was built using rose bengal staining, investigated the different concentrations of luoxianzi extrac- tive on the adherence effect of THP-1 and EA. hy926; the influence of luoxianzai extract on EA. hy926 secreted MCP-1 and VCAM-1 which induced by TNF-α was investigated by ELISA. RESULTS : Luoxianzi extract ( concentration ≤4× 102 μg/mL) had no significant inhibition or proliferation effect in EA. hy926(P〉0.05); lu-oxianzi extract (concentration ≥6.25 /μ/mL) could concentration-dependent decrease the ad- hesion between two cells; all concentrations of luoxianzi extract could significantly reduce the MCP-1 expression induced by TNF-α(P〈 0.01), luoxianzi extract (concentration ≥50 μg/mL), significantly reduced the VCAM-1 ex- pression induced by TNF-a. CONCLUSION: Lu- oxianzi extract can reduce TNF-α-induced endo- thelial cells to secrete MCP-1 and VCAM-1, blocking monocyte adherence to vascular endo- thelial cells, and thus play the role of anti-ather- osclerosis.
