水稻悬浮细胞培养过程中3个逆转座子的转录与转座特性

摘  要:【目的】在水稻悬浮细胞培养不同时期研究3个具有多逆境应答特征逆转座子相关基因的转录与转座特性,为揭示该类基因的生物学功能奠定理论基础。【方法】以水稻品种月亮谷为材料进行悬浮细胞培养,以实时荧光定量PCR检测不同培养时期材料的3个逆转座子相关基因Tos17、Os05g24920和Os01g02040相对表达量及拷贝数的变化,初步分析其转录活性与转座活性。【结果】在培养期间,Os01g02040、Os05g24920、Tos17转录活性呈先升高后下降的变化趋势,其最高转录活性分别出现在培养期的第3个月和第6个月,相对表达量分别为71.84、41.26和53.20倍;Tos17保持较稳定的转录活性,其他2个基因在组培6个月后转录活性迅速下降。在培养3个月后,Os01g02040和Os05g24920的转座活性变化较稳定,相对拷贝数变化范围为2.14-2.64和2.76-5.03倍;Tos17转座活性变化极明显,拷贝数不断提高,至第14个月达12.13倍,明显高于其他两个基因。【结论】在细胞悬浮培养过程中3个逆转座子相关基因均被强烈激活进行转录,但其转录调控和转座调控途径不同。【Objective】The present experiment was conducted to study transcription and transposition characteristics of 3 retrotransposon-related genes responding to different stresses at different stages of rice cell suspension culture in order to provide basis for revealing their biological functions. 【Method】Rice variety Acuche was used as experimental material to conduct cell suspension culture. The changes in relative expression value and copy number of Os01g02040, Os05g24920 and Tos17 at different stages of rice cell suspension culture were detected by real-time fluorescence quantitative PCR to analyze their transcription and transposition activities. 【Result】In the process of culture, the transcription activities of Os01g02040, Os05g24920 and Tos17 presented change trend of increasing firstly and decreasing at later. The highest transcription activity of them was 71.84, 41.26 and 53.20 times when cultured for 3 and 6 months, respectively. Tos17 could keep stable transcription activity, while the others showed sharply decreasing after culturing for 6 months. After 3 months of culture, Os01g02040 and Os05g24920 kept stable transposition activity with 2.14-2.64 and 2.76-5.03 times of relative copy number, respectively. While Tos17 had significant change in transposition activity, its copy number increased continuously and reached 12.13 times on the 14 thmonth,which was higher than that of the other two genes. 【Conclusion】The three genes were all strongly activated to transcribe during cell suspension culture, but their transcription and transposition were regulated in different ways.
