摘  要:目的从茅苍术分离纯化出多糖APW1a,研究其组成及初步结构。方法茅苍术根茎经水提醇沉、脱蛋白、透析,DEAE纤维素与Sephadex G-100凝胶柱洗脱得APW1a;采用紫外、高效凝胶液相、气相、红外、核磁共振等色谱分析其组成及初步结构。结果 APW1a由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖、半乳糖组成,物质的量比为0.16∶0.17∶0.24∶0.55∶0.89∶1分子量约为5.3×103D,糖环以β型吡喃糖为主,并有甲基、酯酰等基团。结论分析了茅苍术多糖APW1a的组成及初步结构,为茅苍术多糖的进一步的研究提供了基础。OBJECTIVE To study the isolation ,the composition and preliminary characterization of polysaccha-ride APW1a from Atractylodes lancea(Thunb.) DC.METHODS The crude polysaccharides were obtained by ex-tracting with boiling-water and precipitating with alcohol ,the protein was removed by TCA ,The cleared polysaccha-ride were purified by DEAE-cellulose,Sephadex G-100 column,received APW1a.The composition and characteriza-tion of APW1a was analyzed by UV,HPLC,GC,IR,1H-NMR.RESULTS APW1a consistsed of Rha、Ara、Xyl、Man、Glu、Gal in a molar ratio of 0.16∶0.17∶0.24∶0.55∶0.89∶1.The molecular mass was 5.3 ×10 3 D.The structure was analyzed by chromatography suggesting APW 1a was in the form of pyranose β-glycosidic bond mainly ,and it con-tained methyl group,ester acyl group and so on.CONCLUSION The structure of APW1a is first discussed,these work cuold establish foundation for the further study of Atractylodes Polysaccharides.
