摘 要:目的:建立测定清热地黄酊中毛蕊花糖苷含量的方法。方法:采用高效液相色谱法。色谱柱为Waters Symmetry Shield C18(250 mm×4.6 mm,5μl),流动相为乙腈-0.1%醋酸溶液(10∶90,V/V),检测波长为334 nm,流速为1.0 ml/min,柱温为30℃,进样量为10μl。结果:毛蕊花糖苷质量浓度在187.6-938.0μg/ml范围内与峰面积积分值呈良好的线性关系(r=0.9997);精密度、稳定性、重复性试验的RSD均≤0.52%;平均加样回收率为99.97%,RSD=0.28%(n=9)。结论:该方法简单、准确、重复性好,可用于清热地黄酊中毛蕊花糖苷的含量测定。OBJECTIVE:To establish the method for the content determination of verbascoside in Qingre dihuang tincture.METHODS:HPLC method was adopted. The determination was performed on Waters Symmetry Shield C18(250 mm×4.6 mm,5 μl)column with mobile phase consisted of acetonitrile-0.1% acetum solution(10∶90,V/V)at the flow rate of 1.0 ml/min. The detection wavelength was set at 334 nm and the column temperature was 30 ℃. The sample size was 10 μl. RESULTS:The linear range of verbascoside was 187.6-938.0 μg/ml(r=0.999 7). RSDs of precision,stability and reproducibility tests were all lower than0.52% with an average recovery of 99.97%(RSD=0.28%,n=9). CONCLUSIONS:The method is simple,accurate and reproducible,and can be used for content determination of verbascoside in Qingre dihuang tincture.
