摘 要:目的建立薄层色谱法和高效液相法监测天然牛黄中是否含有猪去氧胆酸。方法 TLC法采用硅胶G薄层板,以正己烷-乙酸乙酯-醋酸-甲醇(20∶25∶2∶3)的上层溶液为展开剂展开。HPLC法采用Lichrospher C18(4.6mm×250mm,5μm)色谱柱,以乙腈-水-磷酸(35∶65∶0.1)为流动相,检测波长192nm,流速1.0mL·min-1。结果猪去氧胆酸在0.102~5.088mg·mL-1范围内呈良好的线性关系(r=0.9999);加样回收率为97.1%,RSD%为1.2%。结论该法简便准确,灵敏度高,通用性好,适用于牛黄中猪去氧胆酸的监控。OBJECTIVE To establish a TLC and HPLC method for monitoring the hyodeoxycholic acid in calculus bovis. METHODS Silica G was used and developed with n-hexane-ethyl acetate-36 acetic acid-methanol( 20∶25∶2∶3) in the upper solution. A Lichrospher C18( 4.6mm ×250mm,5μm) was used as the analytical column and mobile phase was acetonitrile-water-H3PO4( 35∶65∶0.1) at a flow rate of 1.0mL·min- 1. The UV detection wavelength was set at 192 nm. RESULTS The hyodeoxycholic acid showed a good linear relationship at a range of 0. 102~ 5. 088mg·mL- 1( r =0. 9999); the average recovery was 97.1%,RSD was 1.2%. CONCULSION The method is simple,rapid and reliable for the identification of hyodeoxycholic acid in natural calculusbovis.
