摘  要:目的观测红景天苷对胃癌细胞SGC7901增殖和PKC-α表达的影响,为红景天苷治疗恶性肿瘤提供实验依据。方法采用细胞形态学方法及MTT法、流式细胞术检测分析不同浓度红景天苷对胃癌细胞SGC7901增殖的抑制作用;Western Blot法检测对照组、红景天苷10-40μmol/L各组PKC-α蛋白表达水平。并对上述结果做相关分析。结果红景天苷作用SGC7901细胞24 h后细胞变圆,贴壁生长状态变差,死亡细胞数增多。MTT示红景天苷浓度在10-100μmol/L时,细胞死亡率随药物浓度增加而增多,细胞生存率与红景天浓度呈负相关。流式细胞术结果显示,红景天干预后凋亡细胞随着药物浓度增加而增多(P〈0.05)。Western blot检测结果示PKC-α蛋白的表达随红景天苷浓度(10、20、40μmol/L)的增加逐渐降低(P〈0.05)。结论红景天苷对胃癌细胞SGC7901具有抑制增殖并诱导凋亡的作用。红景天苷对SGC7901细胞有抑制PKC-α蛋白表达的作用。红景天苷对恶性肿瘤具有相关治疗作用。Objective To investigate the proliferation of SGC7901 cells and the expression of PKC- α treated with salidroside and provide the experimental basis for treatment of malignant tumor. Methods To observe morphology of SGC7901 cell and detect inhibition effect in different concentration of salidroside. To analyze the suppressive effects of different concentration salidroside on proliferation of SGC7901 cells and PKC- α expression with MTT method,flow cytometry and Western Blot. Results SGC7901 cells became round and showing a poor adherent growth after 24 h treatment with salidroside. The number of death cells increased. The results of MTT showed that the trend of death cells increased with salidroside concentration in 10 ~ 100 μ mol /L and cell survival rate decreased logarithmically. Flow cytometry showed SGC7901 cells apoptosis and positively correlated with salidroside concentration( P〈0.05). Western blot testing showed that PKC-α expression was negatively correlated with salidroside concentration( 10,20,40μmol /L)( P〈0. 05). Conclusion Salidroside can inhibit the proliferation and induce the apoptosis of SGC7901 cells. Salidroside inhibits PKC- α expression of SGC7901 cells as well. Salidroside may be the strategy of malignant tumor treatment.
