柴芍承气汤对大鼠肠缺血再灌注肠组织半胱氨酰白三烯受体1表达和病理的影响

摘　　要：目的:研究中药柴芍承气汤对大鼠肠缺血再灌注(I/R)保护作用和机理。方法:30只大鼠分为3组,假手术对照组、I/R模型组和中药治疗组各10只。通过免疫组织化学染色方法和RT-PCR法检测大鼠肠组织CysLTR1蛋白和mRNA表达水平,并应用光学显微镜观察小肠病理改变。结果:1与对照组比较,中药治疗组大鼠肠道组织的病理改变均明显减轻。2肠I/R时肠组织CysLTR1蛋白和mRNA表达水平升高;应用中药后,肠组织CysLTR1蛋白和mRNA表达水平均减低。结论:柴芍承气汤能减轻肠I/R肠道组织的病理损害,并能抑制大鼠肠黏膜抑制肠组织CysLTR1的表达,CysLTR1的表达可能是柴芍承气汤保护肠黏膜损伤的靶点。Objective：To study the protective mechanism of Chaishao Chengqi Decoction in rats with intestinal ischemia- reperfusion（I /R）. Methods：30 rats were divided into 3 groups,sham operation group,I /R model group and treatment group with 10 rats in each group. By immunohistochemical staining method and RT- PCR method,we detected the expression of CysLTR1 protein and mRNA levels in rat intestinal tissue,and the optical microscope was used to detect the pathological changes of small intestine. Results：1Compared with the control group,pathological changes of intestinal tissue in rats in the treatment of traditional Chinese medicine were decreased significantly. 2The levels of CysLTR1 protein and mRNA expression in rats intestinal tissue with intestinal I /R were increased. With the application of traditional Chinese medicine,the levels of CysLTR1 protein and mRNA expression in intestinal tissue were decreased. Conclusion：It suggested that the Chaishao Chengqi Decoction could release the pathological change of enteric structure in rat with I /R and Chaishao Chengqi Decoction can inhibit the expression of CysLTR1 in the intestinal mucosa of rats. CysLTR1 may be protective targets of intestinal mucosal injury by Chaishao Chengqi Decoction.
