摘  要:目的:研究痰热清注射液对百草枯(PQ)中毒大鼠肺组织内炎症因子的影响。方法:将72只SD大鼠随机分为生理盐水(NS)对照组、PQ模型组和痰热清干预组,每组24只。采用一次性腹腔注射PQ溶液的方法,制备PQ中毒ALI模型,PQ溶液20mg/kg。PQ模型组和痰热清干预组注射PQ溶液,NS对照组腹腔注射等容积NS。染毒1 h后,痰热清干预组尾静脉注射痰热清1 mL,NS对照组、PQ模型组尾静脉注射等容生理盐水,每24h注射1次。分别在染毒后6 h、24 h、48 h和72 h麻醉处死大鼠,测定肺湿/干重比,进行肺组织HE染色,病理学观察,对肺组织中肿瘤坏死因子-α(TNF-α)和血清中白介素-6(IL-6)含量进行测定。结果:各时间点观察,病理学表明,PQ模型组肺组织水肿、出血和大量的炎性细胞浸润。痰热清干预组的肺组织损伤明显减轻,肺泡腔及支气管腔炎细胞及渗出物与PQ模型组相比明显减少。PQ模型组肺湿/干重比与NS组相比明显增加(P<0.05),肺组织匀浆中TNF-α含量、IL-6含量显著增加(P<0.05);而在痰热清干预组中,肺湿/干重比、肺组织匀浆中TNF-α含量、IL-6含量与PQ模型组相比均明显减少(P<0.05)。结论:痰热清注射液能抑制PQ中毒大鼠肺内炎症因子的表达,减轻肺损伤程度。Objective:To observe the effects of Tanreqing Injection on inflammatory factor in lung tissue of Paraquat poisoning rats. Methods Seventy- two male SD rats were randomly divided into three groups(each n = 24):control group,model group,Tanreqing intervention group. The ALI model induced by PQ poisoning was produced by intraperitoneal injection PQ solution(20 mg /kg),and the rats in control group were given normal saline solution with the same volume.One hour after the administration,rats in Tanreqing intervention group were given Tanreqing Injection,and PQ group were given normal saline solution with the same volume,and injected once every 24 h. At 6,24,48 and 72 hours,the rats were sacrificed,and the lungs were taken away to determine the lung wet /dry weight ratios(W /D ratios) as well as the levels of tumor necrosis factor- α( TNF- α) and interleukin- 6( IL- 6) in the various groups. Results Histological study showed congestion,edema and sequestration of inflammatory cells in the lung tissues in PQ group,but the lung injury was significantly alleviated in Tanreqing intervention group. The W /D ratios of lungs in rats of model group at 6,24,48,72 hours after poisoning were significantly higher than those in the normal control group,and compared with the model group,they were decreased significantly in the Tanreqing intervention group. The levels of TNF- α and IL- 6 in lung tissue of rats in model group at 6,24,48,72 hours after poisoning were significantly higher than those in normal control group,and compared with model group,the level of TNF- α was markedly lower at 6,24,48,72 hours in Tanreqing intervention group(all P〈0. 05). Conclusion Tanreqing Injection could reduce the injury of lung tissue in PQ poisoning rats by inhibiting inflammatory factor.
