摘  要:为了解奶牛乳腺炎病例乳汁中白细胞介素-6(IL-6)与体细胞凋亡的相关性,探讨了患有不同程度乳腺炎的奶牛乳汁中IL-6水平变化,并研究了IL-6与乳汁体细胞凋亡的相关性。利用无菌消毒法对某奶牛饲养场的82头奶牛采集乳样,采用放射免疫分析法测定乳清IL-6,应用流式细胞仪检测乳汁体细胞凋亡率。结果表明,隐性乳腺炎乳汁中IL-6水平极显著高于无乳腺炎乳汁中IL-6水平(P<0.01),临床乳腺炎乳汁中IL-6水平极显著高于无乳腺炎乳汁和隐性乳腺炎乳汁中IL-6水平(P<0.01);奶牛乳腺炎病例乳汁中IL-6水平与上皮细胞、巨噬细胞、淋巴细胞和中性粒细胞凋亡呈明显负相关(P<0.01),其中IL-6与中性粒细胞的相关系数接近1或达到1,二者的相关性最明显,IL-6对中性粒细胞凋亡抑制最明显。奶牛乳汁中IL-6水平与奶牛乳腺炎的发病程度密切相关,IL-6不同程度的抑制牛乳汁中体细胞的凋亡过程,IL-6对中性粒细胞的凋亡过程的明显抑制与奶牛乳腺炎有重要关系。In order to understand the correlation between interleukin-6 (IL-6)and somatic cell apoptosis in mastitis cow milk,we studied the level changes of IL-6 in milks from cows with varying degrees of masti-tis,and then the correlation of IL-6 and somatic cell apoptosis.82 samples were collected aseptically,IL-6 was measured using radioimmunoassay and the apoptosis was detected by flow cytometry.The results showed that the IL-6 levels in milks from recessive mastitis cases were significantly higher than those in normal cases (P 〈0.01),the IL-6 levels from dominant mastitis cases were significantly higher than that from recessive mastitis cases and normal cases (P 〈0.01).The IL-6 levels in mastitis cases were signifi-cantly negatively correlated with apoptosis of epithelial cells,macrophages,lymphocytes,and neutrophil (P 〈0.01).The most obvious correlation were found between IL-6 and neutrophils of which the correlation coefficient were closed 1 or up to 1,and IL-6 suppressed the apoptosis of neutrophils most obviously.The IL-6 levels in cow milk is closely related to the degree of mastitis,IL-6 inhibits apoptosis of somatic cells in different degrees,and the apoptosis process inhibition of IL-6 on neutrophils has an important relationship with mastitis.
