灭幽汤对幽门螺杆菌相关性胃炎脾胃湿热证小鼠AQP4、IL-6、IL-8的影响

摘　　要：目的研究灭幽汤对幽门螺杆菌(Hp)相关性胃炎脾胃湿热小鼠水通道蛋白4(AQP4)蛋白、白介素-6、白介素-8(IL-6、IL-8)的相关性,探讨其治疗慢性胃炎的作用机制。方法将70只BALB/c小鼠随机分为对照组、模型组、灭幽汤高浓度组(灭高组)、灭幽汤低浓度组(灭低组)、胃三联组(替硝唑+克拉霉素+枸橼酸铋钾颗粒),每组14只;采用复合因素(肥甘食物+湿热环境+Hp感染)建立BALB/c小鼠Hp相关性胃炎脾胃湿热证模型;造模成功后连续给药14 d,采用免疫组化检测AQP4的表达情况,Elisa检测血清IL-6、IL-8含量。结果与正常对照组比较,模型组AQP4、IL-6、IL-8表达增加(P<0.01);与模型组比较,灭高组、灭低组、胃三联组小鼠AQP4、IL-6、IL-8表达降低,差异显著(P<0.01);灭高组AQP4、IL-6、IL-8低于胃三联组,但差异无统计学意义(P>0.05)。结论灭幽汤可能通过影响AQP4、IL-6、IL-8表达而发挥治疗Hp相关性胃炎脾胃湿热证的作用。Objective： To study the effect of the mechanism of Mieyou Decoction in the expressions of aquaporin 4 （AQP4）. IL-6,IL-8 in the H.Pylori associated gastritis mice with spleen and stomach damp-heat syndrome. Methods： 70 BALB/c mice were randomly divided into five groups including the control group,the model group, the high concentration of Mieyou Decoction group, the low concentrations of Mieyou Decoction group and the gas- tric triad group （Tinidazole,Clarithromycin and Bismuth potassium citrate granules） with 14 rats in each group. The mice of H.Pylori associated gastritis mice of spleen and stomach damp-heat syndrome model were established by composite factor including the fatness food,the hygropyrexia environment and Helicobacter pylori. After the success of modeling,continuous administration 14 d,the AQPS expression was detectd by the immunohistochem- istry and the IL-6 and IL-8 were detected by the Elisa method. Results： Compared with the control group,ex- pressions of AQP4,IL-6,IL-8 were increased significantly （P〈 0.01 ）. Compared with the model group,above in- dicatiors were decreased obviously in the high concentration of Mieyou Decoction group, the low concentrations of Mieyou Decoction group and the gastric triad group （P〈 0.01 ）. However,there were no statistical differences be- tween the high concentration of Mieyou Decoction and the gastrc trial group （P〉 0.05）. Conclusion： Mieyou De- coction may play a role in treatment of chronic gastritis with spleen and stomach damp-heat syndrome through down regulating the expression of AQP4. IL-6, IL-8 expressions.
