摘 要:目的:观察黄连解毒汤有效部位(总生物碱、总黄酮、总环烯醚萜)对局灶性脑缺血大鼠神经元MAP-2、PGP9.5表达及星形胶质细胞活化的影响,探讨其对神经元-胶质细胞网络的调控作用。方法:中动脉栓塞法建立局灶性脑缺血大鼠模型,SPF级雄性SD大鼠随机分为假手术组、模型组、黄连解毒汤水提物组(800mg/kg)、总生物碱组(44mg/kg)、总黄酮组(50mg/kg)、总环烯醚萜组(80mg/kg),每天灌胃给药1次,连续给药7天。免疫荧光染色结合图像分析技术检测脑缺血大鼠梗死灶周围皮层MAP-2、PGP9.5及星形胶质细胞活化标志蛋白GFAP的表达。结果:脑缺血7天大鼠PGP9.5表达下调,GFAP表达增强,差异较假手术组明显(P<0.01);与模型组相比,黄连解毒汤水提取物组大鼠梗死灶周围神经元MAP-2表达明显上调,GFAP表达下调(P<0.01或P<0.05);总生物碱、总黄酮、总环烯醚萜均可明显上调缺血大鼠梗死灶周围皮层MAP-2、PGP9.5表达,较模型组有统计学差异(P<0.01或P<0.05)。总生物碱还可明显下调GFAP表达,较模型组具有显著性差异(P<0.01)。结论:黄连解毒汤有效部位总生物碱可减轻神经元轴突损伤,促进神经元树突重塑,抑制星形胶质细胞异常活化。Objective : To investigate the effect of Huanglian Jiedu decoction (HLJDD) which consists of total al- kaloids, total flavonoids and total iridoids the expression of MAP - 2 and PGP9.5 and the impact of astrocyte activation. Probe the regulatory role of neuron - glial cells network. Methods : Rat models of focal cerebral is- chemia were established. The expressions of MAP - 2, PGP9.5 and GFAP in isehemic penumbra cortex wereobserved by immunofluorescence staining and then analyzed by imaged imagine analysis system. Male Sprague -Dawley rats were randomly divided into sham group, model group, aqueous extract group, total al- kaloids group, total flavonoids group and total iridoids group. The extracts were intragastrically administrated to rats in doses of aqueous extract group (800mg/kg/day), total alkaloids group (44mg/kg/day), total fla- vonoids group (50mg/kg/day), total iridoids group (80mg/kg/day). Administer each treatment group once every 12h and then sacrifice the rats after seven - day continuous treatment. Results: The expressions of PGP9.5 were excessively decreased, and the expressions of GFAP were increased compared with those of the sham group(P 〈0.01 ), while aqueous extract could increase the MAP- 2 expression and decrease the GFAP expression compared with those of the model group( P 〈0.01 or P 〈0.05 ). Total alkaloids, total flavonoids, total iridoids could increase the MAP - 2 and PGP9.5 expressions compared with those of the model group (P 〈 0.01 or P 〈 0.05 ). Total alkaloids could excessively decrease the expression of GFAP compared with those of the model group(P 〈0.01 ). Conclusion: Total alkaloids are inherently capable of reducing the neu- ronal axonal injury, promoting the neuronal dendritic remodeling and inhibiting the abnormal activation of as- trocytes.
