摘 要:目的:优选活络效灵丹中乳香、没药挥发油提取工艺与包合工艺。方法:采用水蒸汽蒸馏法分别提取乳香、没药挥发油,采用β环糊精包合技术包合挥发油,通过正交试验优选最佳工艺。结果:提取时间对乳香和没药挥发油提取率均有显著性影响,浸泡时间、加水量则无显著性影响,优选提取工艺均为不浸泡、6倍量水提取8h;挥发油与β-CD比例对乳香、没药挥发油包合率均具有显著性影响,转速与剪切时间则无显著性影响,优选包合工艺为挥发油与β-CD的投料比分别为乳香1∶8、没药1∶6,转速均为10000rpm,剪切时间均为15min;薄层色谱表明挥发油被有效包合,且包合前后挥发油主要成分未有明显变化。结论:所优选的挥发油提取与包合工艺可靠可行。Abstract:Objective: To optimize the extraction and inclusion processes for volatile oil from Frankincense and Myrrh. Methods: The essential oil was extracted and included by steam distillation and beta- cyclodextrin (β-CD), and their process parameters were optimized with orthogonal experimental design. Results: Unlike the soak period and water addition for extraction, as well as rotate speed and cut time for inclusion, the extrac- tion duration and the use level of β- CD significantly affected the extraction and inclusion efficiencies of vola- tile oil, respectively. Based on the orthogonal design, the optimized process parameters were generated as be-ing extracted for 8 h with 6 times of water (v/w) but no soak before, for getting volatile oil from both of the medicinal materials mentioned above. The volatile oil was then included with β - CD with the selected process as follow: the usages of Olibanum and β -CD were 1:8 (w/w), and 1:6 for Myrrh volatile oil. The mixture was sheared for 15min under 10 000rpm for achieving an inclusion complex with no free volatile oil and no substances changed, according to the analysis with thin - layer chromatography. Conclusion: The optimized extraction and inclusion processes are suitable for volatile oil.
