摘  要:目的探讨聚合人胎盘血红蛋白溶液(PolyPHb)保存过程中加入Vc的抗氧化效果及最适条件。方法以Vc为抗氧化剂,以固定变量的方法逐个研究溶液中Vc的加入量、pH、温度等因素对Vc抑制高铁血红蛋白(MetHb)生成的影响,并考察Vc在控制MetHb的同时对PolyPHb的结构、载氧活性及主要理化质量指标的影响。结果Vc的最适加入量3≤Vc/Hb(摩尔比)≤8,pH7.5~8.0,在4℃保存抗氧化效果更好;特征光谱扫描、圆二色谱分析和养结合特性曲线分析:Vc作用前后PolyPHb的功能基团、高级结构和氧结合特性无明显区别。结论 Vc能有效抑制MetHb生成,并且对PolyPHb的结构和功能没有影响,可作为血红蛋白类血液代用品保存中的抗氧化剂。Objective To investigate the optimal conditions for adding vitamin C (Vc) in the preservation process of PolyPHb and to explore the effects of Vc on PolyPHb's structure,bioactivity and main physiochemical indexes. Methods Using Vitamin C (Vc) as an antioxidation agent in this research, three influential variables of the antioxidant effect were studied ,including the molar ratio of Vc and hemoglobin, pH and temperature. In addition, through spectral analysis, circular dichroism spectra and the oxygen binding curves,the effect of Vc to PolyPHb's structure, bioactivity and main physiochemical indexes were studied. Results The optimal mole ratio(Vc/Hb) of Vc to add into the sample was 3 ~〈 Vc/Hb ~〈8. The opti- mal pH was 7.5 ~ 8.0. Preserving the sample at 4~C was shown to inhibit MetHb formation for a long time. The results of Spectral analysis, CD and oxygen-binding curves showed no differences before and after the addition of Vc into the PolyPHb solution. Conclusion Vc can inhibit the MetHb formation in PolyPHb solution and had no effects on the molecular structure, bioactivity and main physiochemical indexes of PolyPHb,indicating that Vc can be used as an antioxidation agent in the long-term preservation of HBOCs for its distinct antioxidant properties and negative effects on PolyPHb.
