摘 要:目的探讨不同剂量137Csγ射线辐照对机采血小板中细菌灭活效果及对血小板质量的影响。方法使用20、25、30、35、40 Gy剂量的137Csγ射线分别辐照含有(102~103)CFU/mL铜绿假单胞菌、表皮葡萄球菌、肺炎克雷伯菌的机采血小板,检测辐照前后血小板中的活菌数量和理化指标pH、HSR、血小板最大聚集率、CD62p+、Plt、MPV、PDW,并做统计学分析比较。结果血小板经20、25、30、35、40 Gy的137Csγ射线辐照后,铜绿假单胞菌菌落计数均值分别为314、236、226、160、102 CFU/mL;表皮葡萄球菌计数均值分别为1 440、1 330、1 228、1 175、978 CFU/mL;肺炎克雷伯菌计数均值分别为942、860、750、622、450 CFU/mL。辐照后血小板中活菌数量相比辐照前均明显降低(P<0.05)。不同剂量的137Csγ射线辐照前后血小板pH、HSR、血小板最大聚集率、CD62p+、Plt、MPV、PDW)辐照后变化不大(P>0.05)。结论 20~40 Gy的137Csγ射线辐照机采血小板可较好杀灭铜绿假单胞菌、肺炎克雷伯菌和表皮葡萄球菌,而对血小板质量无明显影响。Objective To investigate the efficiency of different doses of ~37 Cs ^-rays irradiation on bacterial inactivation in apheresis platelets and the effects of different doses of irradiation on the quality of apheresis platelets. Methods The apheresis platelets containing 102 -- 103 CFUs per milliliter with Psemtomonas aeruginosa, Staphylococcus epidermidis and Klebsiella pneumonia were irradiated respectively by 137Csγ--rays with doses of 20,25,30,35 and 40 Gy. Before and after irradiation, the numbers of viable bacteria in plateletand physicochemical indexes of platelet (such as pH, HSR, platelet aggregation rate, CD62P + , platelet count, MPV, PDW)were determined and analyzed. Results The numbers of viable bacteria in platelet decreased significantly after irradiation ( P 〈0. 05 ). After irradiation with doses of 20, 25, 30, 35 and 40 Gy, the average numbers of viable bacteria of Pseudomonas aeruginosa were 314,236,226, 160, and 102CFUs per milliliter; those of Staphylococcus epidermidis were 1 440, 1 330, 1 228, 1 175, and 978 CFUs permilliliter; those of Klebsiella pneumonia were 942, 860, 750, 622 and 450 CFUs per milliliter, respectively. There were differences in the physicochemical indexes of plate before and after irradiation by different amounts of dosage, but the differences were not statistically significant. Conelusion 137Csγ--rays irradiation with 20 -40 Gy dosage could sufficiently kill Pseudomonas aeruginosa, Staphylococcus epidermidis and Klebsiella pneumonia, but had no significant effect on the quality of apheresis platelets.
