摘 要:为了观察蒙古口蘑多糖(Mongolia Tricholoma Polysaccharide,MTP)对低密度脂蛋白(LDL)氧化的影响及其在抗内皮细胞凋亡的作用.采用DPPH法检测MTP的清除自由基活性.将LDL分为MTP处理组和未处理组,经Cu2+氧化处理后,ELISA试剂盒检测它们被氧化程度.同时用这两组被氧化的LDL(ox-LDL)刺激血管内皮细胞,用荧光倒置显微镜和流式细胞仪检测细胞凋亡.实验结果显示MTP具有很高的抗氧化活性,MTP可显著抑制LDL的氧化,抑制血管内皮细胞的凋亡和血管内皮的损伤,从而抑制动脉粥样硬化.Effect of Mongolia Tricholoma Polysaccharide (MTP)on oxidative modification of LDL and its anti-apoptosis effect on Vascular Endothelial Cell (VEC) are investigated. Free radical scavenging activity was tested by DPPH method. To detect vascular protecting effect of MTP, human LDL was divided into the control group and treatment group. The treatment group was disposed of MTP,and then both two groups were oxidized by Cu^2+ into oxidized low density lipo- proteins (ox-LDL). The oxidized level of them was determined by ELISA kit. The VEC apoptosis was detected by the Inverted Fluorescence Microscopy (IFM) and the Flow Cytometry (FCM) after stimulated by ox-LDL of two groups respectively. Results show MTP had a high antioxidant activity and can significantly inhibit oxidative modification of LDL, apoptosis of VEC and vascular endotheli- al damage. Thus MTP can decrease the risk of atherosclerosis.
