摘 要:目的研究石莼、网地藻藻际微生物丰度与藻类多糖含量的相关性。方法通过实时荧光定量PCR(Real-time PCR)技术对石莼、网地藻所附着的藻际微生物丰度进行研究(细菌16SrDNA以及真菌18SrDNA丰度)。结果石莼多糖含量、藻际微生物总DNA含量均显著高于网地藻(P〈0.05)。石莼藻际微生物丰度[细菌(1.04~1.42)×10^10拷贝数/g干样,真菌(1.01~1.38)×10^7拷贝数/g干样]显著高于网地藻[细菌(2.05~6.9)×10^8拷贝数/g干样、真菌(2.59~4.79)×10^5拷贝数/g干样](P〈0.01)。典型对应分析(CCA分析)表明石莼、网地藻多糖含量与藻际微生物(细菌、真菌)丰度显著相关。结论石莼、网地藻藻际微生物丰度对多糖含量有一定影响。Objective To observe the characteristics and abundance of microbial communities in different ulvafor monitoring the potential polysaccharide extraction and microbial factors affecting the health of UlvalactucaLand Dictyota dichotoma.Methods A real-time quantitative PCR(polymerase chain reaction)(RTQ-PCR)method was established to quantitatively detect the abundance of microbial abundance.Results the polysaccharide extraction yield and total DNA content of the UlvalactucaL were significantly higher than that of Dictyota dichotoma(P〈0.05).The results showed that the determination ranges of total bacteria and fungi were 1.04×10^10~1.42×10^10 gene copies/g dry sample and 1.01×10^7~1.38×10^7 gene copies/g dry sample of Ulva lactuca L.The abundance of Ulva lactuca L was significantly higher than that of Dictyota dichotoma(bacteria:2.05×10^8~6.9×10^8 gene copies/g dry;fungi:2.59×10^5~4.79×10^5 gene copies/g dry)(P〈0.01).Results of typical correspondence analysis(CCA)showed that polysaccharide fromUlva lactuca Land Dictyota dichotoma were significantly associated with microbial abundance.Conclusion The abundance of microbial communities has certain influence to polysaccharide extraction.
