摘  要:目的探讨乌司他丁(Ulinastatin,UTI)对大鼠肝缺血-再灌注损伤的保护作用以及对白介素-18(IL-18)表达的影响。方法将大鼠随机分为3组,即假手术组、模型组、乌司他丁组。Pringle法制备大鼠肝缺血-再灌注损伤模型,检测血清中ALT、AST、SOD、MDA含量;测定肝组织中SOD、MDA水平。酶联免疫吸附法(ELISA法)测定大鼠血清及肝组织中IL-18含量。结果乌司他丁组血清中ALT、AST、MDA、IL-18水平低于模型组(P<0.05),SOD活性高于模型组(P<0.05);乌司他丁组肝组织中SOD活性高于模型组(P<0.05),MDA含量和IL-18水平低于模型组(P<0.05)。结论乌司他丁对肝缺血-再灌注损伤具有明显的保护作用,并能够干预大鼠肝缺血-再灌注对白介素-18的过度表达。Objective To investigate the protective effects of ulinastatin(UTI) on hepatic ischemia-reperfusion injury(HIRI) in rats and the effect of that on interleukin-18(IL-18) expression.Methods The rats were randomly divided into 3 groups:sham-operated group,ischemia-reperfusion group and ulinastatin group.HIRI model was made by Pringle method.The levels of ALT,AST,SOD and MDA in serum were detected.The levels of SOD and MDA in hepatic tissue were measured.The serum levels of IL-18 in rats and in the hepatic tissue were detected by ELISA.Results(1) In the ulinastatin group,the serum levels of ALT,AST,MDA and IL-18 were less than those in the ischemia-reperfusion group(P〈0.05) and the levels of SOD in serum were higher than those in the ischemia-reperfusion group(P〈0.05).(2) In the ulinastatin group,the levels of SOD in hepatic tissue were higher than those in the ischemia-reperfusion group(P〈0.05) and the levels of MDA,IL-18 in hepatic tissue were less than those in the ischemiareperfusion group(P〈0.05).Conclusion Ulinastatin has significant protective effects on HIRI and interferes the overexpression of IL-18 by the hepatic ischemia-reperfusion in rats.
