摘 要:目的探讨铅暴露对大鼠血管功能的影响及槲皮素的保护作用。方法成年雄性SD大鼠30只,随机分为对照组、醋酸铅组(1 g/L)和醋酸铅(1 g/L)加槲皮素(30 mg/kg)组,每组10只,通过饮水方式染毒8周。应用试剂盒检测血清和血管组织中内皮素-1(ET-1)、一氧化氮(NO)、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)、丙二醛(MDA),乳酸脱氢酶(LDH)的水平变化,并进行病理形态学观察。结果铅染毒组大鼠血清和主动脉组织中NO、i NOS、MDA和LDH水平较对照组均升高,而SOD水平下降;槲皮素组NO、i NOS、MDA水平较铅染毒组均下降,而SOD水平上升,差异有统计学意义,病理形态学未观察到明显变化。结论铅暴露可以导致实验大鼠血管发生氧化损伤,而槲皮素对其所致损伤有一定的保护作用。Objective To investigate the toxic effect of lead on the cardiac function of rats, and explore the protective effects of quercetin. Methods In total of 30 adult male rats were randomly divided into three groups(n=10), namely the control group(distilled water), lead acetate group(1g/L), lead acetate(1g/L) + quercetin(30mg/kg) group. The lead acetate group was exposed to lead acetate for 8 weeks through drinking water. Rats in lead acetate + quercetin group were administrated both with quercetin at a dose of 30 mg /kg through 4 weeks. The levels of SOD, MDA, Na^+K^+-ATPase, Ca^+Mg^2+-ATPase, CK, LDH levels in heart tissue were detected, and the ECG and pathologic changes were tested. Results Compared with control group, the levels of MDA, Ca^+Mg^2+-ATPase, CK, LDH in lead acetate group increased significantly, and the levels of SOD and Na^+K^+-ATPase decreased. Compared with lead acetate group, the levels of MDA, Ca^+Mg^2+-ATPase, CK, LDH in lead acetate + quercetin group decreased significantly and the level of SOD and Na^+K^+-ATPase were increased. Compared with control group, the results of ECG showed that HP in lead acetate group was decreased significantly, while in PR interval and QT interval increased significantly. Compared with lead acetate group,HP in lead acetate + quercetin group was increased significantly, while in PR interval and QT interval decreased significantly.Pathological examination showed myocarditis performance was in rats of lead acetate group, symptoms of rats in lead acetate +quercetin group had significant relief. Conclusion Lead exposure has an impact on dysfunction of the heart, quercetin can be protective for the damages.
